Abstract :
[en] Animal productions are the results of interactions between the genetic potential of the animal, environmental factors and nutrient supplies. These interactions modulate the endocrine system, especially the somatotrope axis. IGFBP-3 is the major carrier of the IGFs (the main growth-promoting peptide) in plasma circulating as part of a 150 kDa ternary complex with one IGF peptide and a single acid-labile subunit. The IGFBP-3 purification would allow the measure of the changes in plasma IGFBP-3 throughout the day and the development and in various physiological situations. We try to purify the bovine IGFBP-3. Firstly, we load 0,5 ml of serum onto an Hiload 16/6 Superdex 200 prep grade column, then onto an ion exchange column and finally onto an IGF-I affinity column. The purification fails perhaps because the sample is to small. Secondly, 5 ml of serum is fractionated by 3 gel filtrations. Between the first and the second filtration, the fractions containing IGF-I binding activity are pooled and concentrated. Before the third gel filtration, the fractions containing this activity are pooled anew, acidified with HCl and incubated at 4 °C for 2 hours. The third gel filtration fractions are pooled and subjected to an heparin-affinity chromatography. The elution pic shows only IGFBP-3 activity. In conclusion, our purified product contains other proteins. But the ternary complex is dissociated, so an IGF-I affinity chromatography would purify the IGFBP-3 from the elution pic of the heparin-affinity column.
[fr] Les performances zootechniques sont sous la dépendance d’influences génétiques, nutritionnelles et environnementales. Celles-ci engendrent des variations hormonales, notamment au sein de l’axe somatotrope. Dans la circulation sanguine, l’IGFBP-3 est la protéine de transport prédominante de l’IGF-I (le principal médiateur hormonal de l’axe somatotrope qui interagit avec la cellule cible). L’IGFBP-3 forme avec l’IGF-I et une sous-unité acide labile un complexe ternaire. La purification de cette protéine permettrait d’étudier les variations de l’IGFBP-3 au cours du temps et en fonction du stade physiologique. Au cours de ce travail, nous avons tenté de purifier l’IGFBP-3 à partir de sérum de bovin. La première série d’essais n’a pas permis de purifier la bIGFBP-3 au moyen de filtration sur gel, chromatographie d’affinité IGF-I et chromatographie échangeuse d’ions compte tenu du trop petit volume d’échantillon de départ : 0,5 ml. La seconde série d’essais consiste en trois filtrations sur gel (Hiload 16/6 Superdex 200 prep grade) consécutives. Une concentration par ultrafiltration du produit intermédiaire est réalisée entre les 2 premières filtrations sur gel. Une acidification de 2 heures est effectuée avant la dernière filtration sur gel. Le matériel obtenu par cette filtration sur gel est utilisé en chromatographie d’affinité héparine, seul le pic lors du décrochage comporte de l’IGFBP-3. En conclusion, nous n’avons pas réussi à purifier l’IGFBP-3 bovine étant donné que notre produit contient encore des protéines contaminantes. Néanmoins, les résultats sont prometteurs, le complexe ternaire est dissocié. Une chromatographie d’affinité IGF-I devrait purifier l’IGFBP-3 à partir de notre produit de purification.
