Abstract :
[en] This paper gives an overview of experimental and theoretical aspects of the ion mobility spectrometry technique coupled to mass spectrometry. These aspects are illustrated by a recent study on the conformation of a DNA sequence capable of forming intramolecular G-quadruplex folds. This sequence is found in the promoter of the gene coding for the c-Myc protein. The comparison between experimental collision cross sections determined by ion mobility spectrometry and theoretical cross sections calculated for several model structures allows
demonstrating that ions of low charge states have conserved the G-quadruplex parallel fold that was present in solution. Mass spectrometry also revealed the presence of a dimer, which conformation was interpreted using ion mobility : this structure consists of two parallel monomers stacked with each other. [fr] Cet article donne un aperçu des aspects théoriques et expérimentaux de la méthode de spectrométrie de mobilité ionique couplée à la spectrométrie de masse (SMI/SM). Ceux-ci sont illustrés par une étude portant sur la conformation adoptée par une séquence d’ADN du promoteur du gène codant pour c-Myc, cette séquence pouvant se replier en une structure de type G-quadruplex. La comparaison entre les sections effi caces de collision déterminées par spectrométrie de mobilité ionique et les sections efficaces calculées pour des structures modèles permet de montrer que les ions d’états de charge les plus faibles gardent en phase gazeuse la conformation en G-quadruplex parallèle qui était présente en solution. La spectrométrie de masse a également permis de révéler la présence d’un dimère et la mise en oeuvre de la spectrométrie de mobilité ionique a conduit à la déduction de sa conformation, cette structure se
trouvant constituée de deux G-quadruplex parallèles empilés.
