Abstract :
[fr] La mélatonine, sécrétée principalement par la glande pinéale pendant la nuit, est l'hormone clé de la régulation de nos rythmes chrono biologiques, tels que les rythmes hormonaux circadiens, le rythme de température et de veille sommeil. Mc Cord et Allen étudient en 1917 les effets des extraits de la glande pinéale sur les têtards et découvrent l'effet éclaircissant de la peau à partir de leur action sur les mélanocytes. En 1958, Aaron Lerner et ses collaborateurs redécouvrent ces expériences, et en utilisant des milliers de glandes pinéales bovines réussissent à isoler la mélatonine. Le prix Nobel de physiologie et/ou Médecine , Julius Axelrod décrira la voie de synthèse de la mélatonine à partir du triptophane, tout en étudiant la riche inervation sympathique de la glande pinéale par le ganglion cervical supérieur.L'ensemble de ces chercheurs étendront ainsi nos connaissances sur la mélatonine. Dans cet article, nous synthétisons les connaissances actuelles sur la mélatonine en tant qu'agent chrono biotique endogène et médicament chrono biotique (Circadin, Ramelteon, Tasimelteon, Agomelatonine). Nous décrivons également les conditions cliniques des états d'hyper et d'hypomélatoninémie chez l'homme. L' anti-radicaux libres in vitro de la mélatonine fait l’objet de nombreuses recherches, qu’il faudra suivre avec attention, notamment dans le cadre de l’oncologie et du vieillissement.
[en] Melatonin, secreted during the night mainly by the pineal gland, is the key hormone for the regulation of our chrono-biological rhythms, such as the circadian hormonal rhythms, the temperature and the sleep-wake rhythm. Mc Cord and Allen studied in 1917 the effects of extracts from the pineal gland on tadpoles and discovered the lightening effect from their action on melanocytes. In 1958, Aaron Lerner and his collaborators rediscovered these experiments, and by using thousands of bovine pineal glands succeed in isolating melatonin. The Nobel Prize in Physiology and / or Medicine, Julius Axelrod will described the synthetic route of melatonin synthesis from triptophan, while studying the rich sympathetic inervation of the pineal gland by the upper cervical ganglion. All of these researchers will extend our knowledge about melatonin. In this article, we synthesize current knowledge on melatonin as an endogenous chrono biotic agent and chrono biotic drug (Circadin, Ramelteon, Tasimelteon, Agomelatonin). We also describe the clinical conditions of hyper and hypomelatoninemia in humans. The in vitro anti-free radicals of melatonin are the subject of recent research, which should be followed carefully, especially in the context of oncology and the aging process.
