Abstract :
[en] Control measures of postharvest fungal diseases of plants are principally based on the use of conventional chemical fungicides. However, in view of the problems associated with the use of these ones, as well as the increasing demand for organically produced food, there is a need to replace chemical fungicides by alternative safer products, such as biological fungicides called "natural" or biofungicides. Among natural fungicides, essential oils (EOs) have been studied. EOs and their compounds present, among other things, antifungal properties and can be an interesting alternative to chemical fungicides. For this purpose, a screening of EOs according to intrinsic (yield in aromatic plant, toxicity) and extrinsic (availability, cost) criteria was carried out. Thirty EOs were selected and their inhibitory activity was tested on the in vitro growth of Penicillium italicum Wehmer, P. digitatum Sacc (pathogens of citrus fruits) and Colletotrichum musae (Berk. & M.A. Curtis) (pathogen of banana). Cinnamomum zeylanicum, C. verum and Eugenia caryophyllata proved to be the most effective at 100 ppm against the three pathogens. The biological activity of these three EOs against P. digitatum and P. italicum was evaluated on artificial infected orange fruits. EOs diluted in ethanol were able to either partially (at a concentration < 5%) or totally (at a concentration ≥ 5%) control disease incidence due to infections of both pathogens. Among the three EOs, C. zeylanicum was the most effective. In order to improve the efficiency of C. zeylanicum EO, a development of formulations consisting of this EO and Simulgel (thickening and emulsifying polymer) or waxes (shellac and / or carnauba, paraffin or polyethylene) were performed. The in vivo biological activity of the formulations was also evaluated against P. digitatum and P. italicum. In general, fungal disease control was improved in the case of fruits treated with C. zeylanicum EO supplemented with Simulgel or waxes compared to that supplemented with ethanol. The improvement was much higher with formulation consisting of C. zeylanicum-shellac (and / or carnauba) compared to the others. A SPME/GC-MS analysis of the headspace of EOs (C. zeylanicum, C. verum and E. caryophyllata) supplemented with shellac and / or carnauba (in vitro) or of oranges treated with such formulations (in vivo) highlighted the lack of some specific volatile compounds of EOs (oxygenated cyclic ones with a specific gravity superior to 0.934), in the vapor phase of the in vitro and in vivo tests, even after 7 days. While when these EOs were supplemented with ethanol, Simulgel, paraffin or polyethylene, all EO compounds were detected in the analyzed headspaces from 4 hours up to 7 days. Analysis of formulations by Fourier transform infrared spectroscopy (FTIR) revealed the formation of hydrogen bonds between such specific EO compounds and waxes (shellac and / or carnauba) This hydrogen bonds formation seems to be responsible of the non-volatilization of EO compounds and their maintaining on the fruit epidermis, thus improving citrus fruit protection. These hydrogen bonds are not observed between non-oxygenated or non-cyclic oxygenated EO compounds and none of the waxes. The present study may therefore allow a careful selection of appropriate EOs and waxes for the elaboration of effective biofungicide formulations. This application could be integrated into the packline procedure without significant modification.
[fr] Les mesures de contrôle des maladies cryptogamiques des plantes en post-récolte sont principalement basées sur l'utilisation de fongicides chimiques conventionnels. Cependant, compte tenu des problèmes liés à l'utilisation de ceux-ci, ainsi qu'à la demande croissante de produits alimentaires biologiques, il y a une nécessité de remplacer les fongicides chimiques par des produits alternatifs plus sûrs, tels que les fongicides biologiques dits «naturels» ou biofongicides. Parmi les fongicides "naturels", les huiles essentielles (HEs) ont été étudiées. Les HEs et leurs composés présentent, entre autres, des propriétés antifongiques et peuvent être une alternative intéressante aux fongicides chimiques. A cet effet, un criblage d'HE, sur la base de critères intrinsèques (rendement, toxicité) et extrinsèques (disponibilité, coût) a été effectué. Trente huiles essentielles ont été sélectionnées et leur activité inhibitrice a été testée sur la croissance in vitro de Penicillium italicum Wehmer, P. digitatum Sacc (agents pathogènes des agrumes) et Colletotrichum musae (Berk. & MA Curtis) (agents pathogènes de la banana). Les HEs de Cinnamomum zeylanicum, C. verum et Eugenia caryophyllata se sont avérées être les plus efficaces à 100 ppm contre les trois agents pathogènes. L'activité biologique de ces trois HEs, contre P. digitatum et P. italicum, a été évaluée sur des oranges infectées artificiellement. Les HEs diluées dans de l'éthanol ont été capable de contrôler partiellement (à une concentration <5%) ou totalement (à une concentration ≥ 5%) l'incidence des maladies dûes aux infections de ces deux agents pathogènes. Parmi les trois HEs, C. zeylanicum était le plus efficace. Afin d'améliorer l'efficacité de l'HE de C. zeylanicum, des formulations constituées de cette HE et de Simulgel (polymère épaississant et émulsifiant) ou de cires (gomme-laque et/ou de carnauba, de paraffine ou de polyéthylène) ont été développées. L'activité biologique in vivo des formulations a également été évaluée contre P. digitatum et P. italicum. En général, le contrôle de la maladie a été amélioré dans le cas des fruits traités par l'HE de C. zeylanicum additionnée de Simulgel ou de cires, par rapport à celle additionnée d'éthanol. L'amélioration était beaucoup plus élevée avec la formulation constituée de C. zeylanicum-gomme laque (et/ou carnauba) par rapport aux autres. Une analyse SPME / GC-MS de l'espace de tête des HEs (C. zeylanicum, C. verum et E. caryophyllata) additionnées de gomme-laque et/ou de carnauba (in vitro); ou d'oranges traitées avec de telles formulations (in vivo), a mis en évidence l'absence de certains composés volatils spécifiques des HEs (composés cycliques oxygénés avec une densité supérieure à 0,934), dans la phase vapeur des tests in vitro et in vivo, même après 7 jours. Alors que, lorsque ces HEs ont été additionnées d'éthanol, Simulgel, paraffine ou de polyéthylène, tous les composés des HEs ont été détectés dans les espaces de tête analysés à partir de 4 heures jusqu’à 7 jours. L'analyse des formulations par spectroscopie infrarouge à transformée de Fourier (IRTF) a révélée la formation de liaisons hydrogène entre ces composés spécifiques des HEs et les cires (gomme-laque et/ou de carnauba). Cette formation de liaisons hydrogène semble responsable de la non-volatilisation des composés des HEs et de leur maintien sur l’épiderme du fruit, améliorant ainsi la protection des agrumes. Ces liaisons hydrogène ne sont pas observées entre les composés non oxygénés (E-caryophyllène) ou les composés oxygénés non cycliques (linalool) des HEs et les cires. La présente étude peut donc permettre une sélection rigoureuse et appropriée des HEs et des cires pour l'élaboration de formulations de biofongicides efficaces. Cette application pourrait être intégrée dans la procédure packline sans procéder à des modifications significatives.
