The atypical chemokine receptor ACKR3/CXCR7 plays crucial roles in numerous physiological processes but also in viral infection and cancer. ACKR3 shows strong propensity for activation and, unlike classical chemokine receptors, can respond to chemokines from both the CXC and CC families as well as to the endogenous peptides BAM22 and adrenomedullin. Moreover, despite belonging to the G protein coupled receptor family, its function appears to be mainly dependent on β-arrestin. ACKR3 has also been shown to continuously cycle between the plasma membrane and the endosomal compartments, suggesting a possible role as a scavenging receptor. So far, the molecular basis accounting for these atypical binding and signalling properties remains elusive. Noteworthy, ACKR3 extracellular domains bear three disulphide bridges. Two of them lie on top of the two main binding subpockets and are conserved among chemokine receptors, and one, specific to ACKR3, forms an intra-N terminus four-residue-loop of so far unknown function. Here, by mutational and functional studies, we examined the impact of the different disulphide bridges for ACKR3 folding, ligand binding and activation. We showed that, in contrast to most classical chemokine receptors, none of the extracellular disulphide bridges was essential for ACKR3 function. However, the disruption of the unique ACKR3 N-terminal loop drastically reduced the binding of CC chemokines whereas it only had a mild impact on CXC chemokine binding. Mutagenesis also uncovered that chemokine and endogenous non-chemokine ligands interact and activate ACKR3 according to distinct binding modes characterized by different transmembrane domain subpocket occupancy and N-terminal loop contribution, with BAM22 mimicking the binding mode of CC chemokine N terminus. The brain has a complex organization portraying multiple topographies at different levels and along different axes. Following the developments of neuroimaging technics in the last decades, different approaches have been developed to study brain organization based on neuroimaging features. Furthermore, many concepts from the psychological sciences have been mapped to the brain. Thousands of fMRI and PET studies show the neural correlates of various concepts derived from the study of human behavior. An overview of those model-based data reveals a many-to-many mapping on both brain-behavior directions: one single brain region or network can be related to many behavioral concepts and conversely, one single behavioral concept can be assigned to many brain regions/networks. The aggregation of these behavioral model-based data can now be harnessed to switch the perspective. By using quantitative approaches on available neuroimaging data, we can derive “behavioural profiling” of brain regions and networks. Such data-driven approaches can be used, in turn, to generate a brain-based model of cognitive information processing. [less ▲]

They range from local properties observed in the brain tissue to long-range connectivity patterns and capture similar, but also complementary dimensions of brain organization. AMPA receptors (AMPARs), one of three sub-groups of ionotropic glutamate receptors present in the central nervous system, are recognised for their involvement in long-term potentiation (LTP), and learning and memory processes.1 They represent a valid cognitive enhancer target, particularly in the fight against Alzheimer’s disease (AD).2,3 Benzothiadiazine 1,1-dioxides, such as BPAM121, have emerged as important allosteric modulators of AMPARs, working solely in the presence of the endogenous transmitter.4 Synthesis of BPAM121 labelled with fluorine-18 was proposed, to investigate the utility of this molecule as a PET probe in vivo, and evaluate its potential as an AD diagnostic tool (Figure 1). Countless technologies and chemical processes make use of nanoporous materials: heterogeneous catalysis, including electrochemical reactions in fuel-cell electrodes, adsorption separation processes, kinetically selective membrane processes, are but a few examples. Small-angle scattering of either x-rays (SAXS) or neutrons (SANS) is one of the few experimental methods available for the in situ analysis of phenomena in this type of materials at the mesoscopic scale. In this presentation, we briefly review some recent applications of small-angle scattering to empty and loaded mesoporous solids [1-3]. A particular focus is put on the data analysis challenges, whereby the scattered intensity is converted to real-space structures with nanometer resolution [4,5]. [less ▲]

Vitamin B1, in the form thiamin diphosphate (ThDP), serves as an enzymatic cofactor for several enzymes involved in glucose metabolism, the Krebs cycle, and branched-chain amino acid biosynthesis. Vitamin B6 in its form as pyridoxal 5'-phosphate (PLP) is required as a cofactor for more than 140 enzymatic reactions in cells, primarily in amino acid, sugar and fatty acid metabolism. Mammals lack the ability to biosynthesize both vitamins B1 and B6 de novo, and plants represent one of their major sources. Symptoms of severe vitamin B1 deficiency include degeneration in the nerves and heart, a disease known as beriberi, which can be fatal. Acute vitamin B6 deficiency can lead to various chronic diseases in humans, including neurological disorders, cardiovascular disease and diabetes. Due to their importance in regions where vitamins B1 and B6 deficiencies are reported, we identified cassava and rice as good target crops for biofortification. In the present work, we performed a screening for B1 and B6 accumulation in selected varieties and landraces representative of the rice and cassava diversity. HPLC analysis allowed precise quantification of vitamin accumulation in selected genotypes contrasting for vitamin accumulation. We also report the first profiling of B1 and B6 vitamers in cassava storage roots and rice endosperm. This work is of primary importance given the reported varying degree of bioavailability for certain vitamers. Identification and expression analysis of B1 and B6 biosynthetic genes from rice and cassava was instrumental to find correlation between expression of selected biosynthetic genes and vitamin accumulation. Genetic engineering of vitamin biosynthetic pathways led to the generation of transgenic rice and cassava lines with high vitamin B6 content. Importantly, our Caco-2 cell assay demonstrated that field-grown storage roots from transgenic cassava lines accumulate high levels of bioavailable vitamin B6. Despite their key role in plant metabolism and their antioxidant activities, increased accumulation of vitamin B6 in transgenic rice and cassava lines could not be associated with improved tolerance to selected biotic or abiotic stresses under lab conditions. Genetic engineering of vitamin biosynthetic pathways led to the generation of transgenic rice and cassava lines with high vitamin B6 content. Importantly, our Caco-2 cell assay demonstrated that field-grown storage roots from transgenic cassava lines accumulate high levels of bioavailable vitamin B6. Despite their key role in plant metabolism and their antioxidant activities, increased accumulation of vitamin B6 in transgenic rice and cassava lines could not be associated with improved tolerance to selected biotic or abiotic stresses under lab conditions. De novo protein design is a growing field in protein chemistry, where artificial proteins are first designed in silico and then validated experimentally. Our group has a long tradition in the design of artificial (β/α)8-barrel proteins, known as Octarellins [1]. This fold, also known as TIM-barrel, is widespread in nature, particularly in enzymes, and represent an interesting target for therapeutic or biological applications. Here we present a novel protocol for the de novo design of TIM-barrels. Both Rosetta and Modeller modeling softwares were used to create the backbone structure of a TIM-barrel fold and to generate more than 10.000 artificial amino acid sequences. Stability was tested for the more interesting models by running molecular dynamics (MD) simulations, using GROMACS. The best models were chosen for protein production and preliminary biophysical characterization. The design of artificial proteins and the improvement of bioinformatics tools for protein modeling, structure prediction and MD simulations seem to be essential for a comprehensive knowledge of protein structure in general, as well as for an optimal use of the massive amount of data resulting from the numerous genome sequencing projects. Cassava Mosaic Disease (CMD) is a major constraint to cassava production in Africa and in the Indian sub-continent. CMD is caused by cassava geminiviruses (CGMs), which are bipartite ssDNA viruses. At least 9 CGM species are infecting cassava and CGM diversity is particularly high on the African continent. In order to engineer resistance against African cassava mosaic virus (ACMV), we developed transgenic cassava expressing dsRNA against the ACMV Rep (AC1) sequences that are conserved amongst CGMs. Transgenic dsAC1 cassava lines were selected based on the accumulation of transgene-derived small RNAs. We established a rapid and high-throughput agroinoculation method in order to assess virus resistance in cassava. Selected transgenic cassava lines displayed robust resistance against increasing loads of ACMV delivered by gene gun and by agroinoculation. We also observed a fast recovery phenotype in transgenic lines following graft inoculation of ACMV. Best performing dsAC1 cassava lines were also inoculated with South African cassava mosaic virus (SACMV) and East African cassava mosaic virus (EACMV). Recovery from SACMV and EACMV infections in transgenic cassava lines was delayed as compared to the recovery phenotype observed with ACMV infection. Collectively, our results suggest that high similarity between transgene-derived dsRNAs and target virus sequences is required to engineer transgenic plants rapidly recovering from virus infection. Selected transgenic dsAC1 cassava lines also showed delayed development of CMD symptoms in the field. Molecular analysis of CGM species in the field is being performed in order to better characterize the level of resistance of dsAC1 lines against multiple CGM species. [less ▲]
