It is here demonstrated that it is possible to completely restore sound-induced pexophagy and to prevent oxidative stress in Pjvk-/- auditory hair cells by transducing in same the Pejvakin and MAP1LC3B proteins together. According to the present invention a polyvinyl alcohol is produced by performing: a polymerization step wherein a polymer solution containing a polyvinyl ester is obtained by polymerizing a vinyl ester monomer by means of controlled radical polymerization in the presence of a radical initiator and an organic cobalt complex; an extraction step wherein a cobalt complex is extracted from the polymer solution by bringing the polymer solution into contact with an aqueous solution containing a water-soluble ligand; and a saponification step wherein a polyvinyl alcohol is obtained by saponifying the polyvinyl ester after the extraction step. Consequently, the present invention provides a method for producing a polyvinyl alcohol which has narrow molecular weight distribution, high number average molecular weight and good hue, while exhibiting good solubility in water. MT4-MMP (MMP-17) is a glycosylphosphatidyl inositol (GPI) anchored MMP produced by cancer cells that promotes tumor vascularization and metastases. The present invention found that MT4-MMP expression promotes cancer cell proliferation. These effects involve retinoblastoma protein (Rb) inactivation, cyclin dependent kinase CDK4 activation and Epidermal Growth Factor Receptor (EGFR) signaling. Co-immuno-precipitations indicate the existence of protein complexes harboring MT4-MMP and EGFR. The present invention further found a novel mechanism of MT4-MMP action through an outside-in signaling involving EGFR. An unexpected crosstalk between an MMP and EGFR was identified and recognized as a key driver of cancer cell biology. [less ▲]

The present invention relates to an implant comprising: - a core material comprising polydimethylsiloxane or at least one hydrogel polymer; - a tube encasing said core material comprising an ethylene vinyl acetate polymer or at least one hydrogel polymer; - a sealant for closure of the open ends of said tube comprising polydimethylsiloxane or a mono-, di-, or triacetoxy derivative thereof, or at least one hydrogel polymer; and - at least one active ingredient; wherein said at least one active ingredient is selected from the group comprising celecoxib, sulindac, tamoxifen, oestrogen, oestradiol, ethinyl oestradiol, mestranol, dienogest, norgestrel, levonorgestrel, desogestrel, norgestimate, ethynodiol diacetate, leuprorelin, buserelin, gonrelin, triptorelin, nafarelin, deslorelin, histrelin, and supprelin; and with the proviso that when the sealant is said at least one hydrogelpolymer, the core material comprises polydimethylsiloxane. Furthermore, the invention relates to an implant for use as a medicament. In particular, the invention relates to an implant for use in the treatment of endometriosis. [less ▲]
