Equine atypical myopathy (AM) is caused by hypoglycin A (HGA) and methylenecyclopropylglycine (MCPG) intoxication resulting from the ingestion of seeds or seedlings of some Acer tree species. Interestingly, not all horses pasturing in the same toxic environment develop signs of the disease. In other species, it has been shown that the intestinal microbiota has an impact on digestion, metabolism, immune stimulation and protection from disease. The objective of this study was to characterize and compare fecal microbiota of horses suffering from AM and healthy co-grazers. Furthermore, potential differences in fecal microbiota regarding the outcome of diseased animals were assessed. This prospective observational study included 59 horses with AM (29 survivors and 30 nonsurvivors) referred to three Belgian equine hospitals and 26 clinically healthy co-grazers simultaneously sharing contaminated pastures during spring and autumn outbreak periods. Fresh fecal samples (rectal or within 30min of defecation) were obtained from all horses and bacterial taxonomy profiling obtained by 16S amplicon sequencing was used to identify differentially distributed bacterial taxa between AM-affected horses and healthy co-grazers. Fecal microbial diversity and evenness were significantly (p < 0.001) higher in AM-affected horses as compared with their non-affected co-grazers. The relative abundance of families Ruminococcaceae, Christensenellaceae and Akkermansiaceae were higher (p ≤ 0.001) whereas those of the Lachnospiraceae (p = 0.0053), Bacteroidales (p < 0.0001) and Clostridiales (p = 0.0402) were lower in horses with AM, especially in those with a poor prognosis. While significant shifts were observed, it is still unclear whether they result from the disease or might be involved in the onset of disease pathogenesis. [less ▲]

Equine atypical myopathy (AM) is seasonal intoxication resulting from the ingestion of seeds and seedlings of the sycamore maple (Acer pseudoplatanus) that contain toxins, among them, hypoglycin A (HGA). Literature mentions several cases of AM among gravid mares and in unweaned foals. The objective of this study was to determine whether HGA and/or its metabolite are present in milk from grazing mares exposed to sycamore maple trees as confirmed by detection of HGA and its metabolite in their blood. Four mare/foal couples were included in the study. Both HGA and its metabolite were detectable in all but one of the milk samples. To our knowledge, this is the first study describing transfer of HGA to the milk. This unprecedented observation could partially explain cases of unweaned foals suffering from AM. However, a transplacental transfer of the toxin cannot be excluded for newborn foals. Besides being a source of contamination for offspring, milk contamination by toxins from fruits of trees of the Sapindaceae family might constitute a potential risk for food safety regarding other species' raw milk or dairy products. Atypical myopathy (AM) is a severe and often fatal muscle disorder arising from ingestion of toxins contained in sycamore maple (Acer pseudoplatanus) seeds and seedlings. Progresses in research to diagnose and treat AM after intoxication are moving forward, but the acuteness of the disease with a fatality rate of ~75% reinforces the need for preventive measures. The condition is a seasonal disorder. Ingestions of Samaras (seeds) in autumn and seedlings the following spring causes two subsequent clinical case series, with a majority of cases observed in autumn. [less ▲]

In 2014, atypical myopathy (AM) was linked to Acer pseudoplatanus (sycamore maple) in Europe. The emergence of this seasonal intoxication caused by a native tree has raised many questions. This manuscript aims at answering the five most frequently asked questions (FAQs) regarding (1) identification of toxic trees, reduction of risk at the level of (2) pastures and (3) equids, (4) the risk associated with pastures with sycamores that have always been used without horses being poisoned and (5) the length of the risk periods. Answers were found in a literature review and data gathered by AM surveillance networks. A guide is offered FAQ1: a guide is offered to differentiate common maple trees (FAQ1). In order to reduce the risk of AM at pasture levell: (FAQ2): avoid humid pastures, permanent pasturing, spreading of manure and for pasture with sycamores in the vicinity and avoid sycamore maple trees around pasture (FAQ2). To reduce the risk of AM and, at horse level (FAQ3 ): during risk periods, reduce pasturing time according to weather conditions and to less than six hours a day during risk periods for horses on risk pasture, provide supplementary feeds including toxin-free hay, water from the distribution network, vitamins and a salt block (FAQ3). AFAQ4: all pastures with a sycamore tree in the vicinity are at risk (FAQ4). FAQ5: Ninety-four 94% of cases occur over a two 3-month periods, starting in October and in March, for cases resulting from seeds and seedlings ingestion, respectively (FAQ5). [less ▲]

Since mitochondria play diverse crucial roles within the cell, mitochondrial dysfunction can have disastrous consequences. High resolution respirometry (HRR) is a new tool that allows the quantification of mitochondrial oxidative phosphorylation and electron transfer capacities. This tool allows an analysis of mitochondrial bioenergetics and the main substrate pathway of the cells that are of interest. In human, platelets and peripheral blood mononuclear cells have been used as biomarkers in diverse pathological models, as diabetes, atherosclerosis, cancer and neurodegenerative diseases. In the horse, HRR studies have been conducted on skeletal muscles samples. Using this technique, a so-called “mitochondrial syndrome” has been reported in equine atypical myopathy, an environmental intoxication, which is characterized by a severe decrease of mitochondrial respiratory capacity in affected individuals. However, the muscle biopsy procedure is difficult to use in equine clinical routine. Therefore, the aim of this study was to test whether mitochondrial function could be assessed on equine blood mononuclear cells with respirometry. Whole blood was collected in 9 ml EDTA tubes in six horses. Cells of interest (PBMC) were isolated with a density-separation medium (Lymphoprep™) following a standardized procedure. Electron paramagnetic resonance as well as HRR analysis were performed within the following 6 hours. With this isolation technique more than 1x106 PBMC/ml of whole blood were obtained. Respirometric analysis with PBMC taken from the same horses indicated a lack of reproducibility. This finding seems to be primarily due to a too long timeframe (i.e. ±Xh) between cell isolation and respirometric analysis. Additional studies indicated that when respirometry was performed within the first 2 hours after cell isolation, standard variation between samples was significantly reduced. Up to now a correlation between skeletal muscle fiber and PBMC mitochondrial bioenergetics in the equine species is not possible to be established. A standardization of the procedure is vital for reproducibility between laboratories and comparison between healthy and ill horses. The next steps are to compare the oxygraphic measures between equine PBMC and muscle biopsies, as well as the comparison of mitochondrial function between healthy and horses in extreme metabolic conditions (e.g atypical myopathy). Atypische Weidemyopathie ist eine bei Equiden bekannte Erkrankung, die durch die Aufnahme von toxinhaltigen Anteilen des Bergahorns (Acer pseudoplatanus) ausgelöst wird. Das Toxin Hypoglycin A ist in den Samen und Sämlingen von Bergahorn-Pflanzen in besonders hoher Konzentration enthalten; entsprechend tritt die Erkrankung vor allem im Herbst und im Frühling auf, wenn die jeweiligen Pflanzenteile für Tiere erreichbar sind. Hypoglycin A wird zu MCPA-CoA metabolisiert, welches die Fettsäureoxidation in den Mitochondrien hemmt. In der Folge kommt es zu Energiemangel in lebenswichtigen Organen (Herz, Hirn, Atemmuskulatur) und einer massiven Rhabdomyolyse. Erkrankte Pferde zeigen Steifheit, Schwäche und Myoglobinurie, die Mortalitätsrate liegt bei 70-90%. 1 [less ▲]

An adaptive capacity of white adipose tissues is essential to match energy intake and expenditure. Part of this adaptation is driven by the enrollment of new adipocytes, a phenomenon referred to “adipogenesis”. Our current efforts are dedicated to the investigation of the putative role of the Sirtuin 3 (SIRT3), a NAD+-dependent deacetylase and master regulator of several mitochondrial functions, in the differentiation of preadipocytes in white adipocytes. This is of particular relevance as mitochondrial biogenesis and activation of oxidative metabolism are promoted by SIRT3 and are known to be required for proper adipogenic differentiation. Moreover, SIRT3 was already reported to be required for proper differentiation of various other cell types such as skeletal myocytes [1] and brown adipocytes [2]. White 3T3-L1 adipoblasts were differentiated in adipocytes in the presence or in the absence of SIRT3 (invalidation using CIRSPR/Cas9-DN technology) and the expression of SIRT3 as well as markers of its activity were analysed during a differentiation programme of 12 days. In addition, ROS production as well as respiration (Oxygraph-2k, ORBOROS Instruments Corp) in cells in the presence or in the absence of SIRT3 were studied. The results show that the expression of SIRT3 is induced during the differentiation programme. In addition, the absence of SIRT3 is reflected by a delay (but not the inhibition) of preadipocyte differentiation as demonstrated by a reduced content of triacylglycerols. We are currently investigating the underlying mechanisms that could postpone adipogenesis in the absence of the mitochondrial deacetylase such as modifications in reactive oxygen species production and putative alterations of OXPHOS that could connect SIRT3 to proper development of the adipose phenotype. In conclusion, we showed that SIRT3 activity accompanies the adipogenic programme of 3T3-L1 preadipocytes and that the enzyme plays an important role in the kinetics of the differentiation programme. The identification of SIRT-3 dependent mechanisms playing a role in the differentiation of preadipocytes is ongoing. [less ▲]
