Objectifs : Le contrôle de la pression artérielle (PA) chez le patient hémodialysé (HD) est un enjeu majeur en raison des résultats controversés sur les risques cardiovasculaires qui y sont associés. Cependant, la mesure de la PA dans cette population est difficile à appréhender au vu de sa grande variabilité. L’exactitude des mesures péridialytiques (enregistrées dans l’unité d’HD pendant la séance) est souvent remise en question. L’objectif de notre étude est d’établir la contribution et la concordance entre deux techniques de mesure ambulatoire pour détecter la PA non contrôlée chez ces patients HD. Il s’agit du monitoring ambulatoire interdialytique de 44h (MAPA) et de l’automesure au domicile (HBPM) pendant une durée variable de 3, 5 et 7 jours. Méthodologie : 43 patients hémodialysés chroniques (H=28, F = 15), avec une moyenne de 68,3±13 ans ont réalisé une MAPA de 44h (Spacelabs 90207), de la fin d’une séance d’HD au début de la suivante, immédiatement suivie par une automesure de 7 jours (Omron M6). Les mesures péridialytiques étaient la moyenne des PA pré-dialyse ou post-dialyse enregistrées sur 2 semaines (6 séances d’HD). L’hypertension était définie par une PA égale ou supérieure à 140/90 mmHg pour la PA pré-HD, 130/80 mmHg pour la PA post-HD, 130/80 mmHg pour la MAPA de 44 h et 135/85 mmHg pour l’HBPM. Résultats : Nous avons constaté de bonnes corrélations entre la MAPA et l’HBPM pour le diagnostic de l’hypertension chez le patient HD. Comparées aux PA péridialytiques, les 2 techniques ambulatoires ont permis d’identifier la même proportion d’hypertension masquée et d’hypertension de la blouse blanche (25%). La plus grande précision de diagnostic sur le status tensionnel est retrouvée pour les plus longues périodes d’enregistrement (44h pour la MAPA et 7 jours pour l’HBPM). Les plus courtes périodes étaient mieux tolérées par les patients mais moins précises pour détecter l’hypertension. L’automesure était plus appréciée que la MAPA de 44h. Conclusion : La MAPA et l’automesure ont permis d’identifier un quart de patients avec un phénotype particulier d’hypertension, qui n’aurait pas été classé correctement en se fiant uniquement aux mesures péridialytiques. L’automesure devrait être recommandée en première ligne chez le patient HD (au moins une semaine/mois). Cependant, la MAPA fournit des informations uniques comme la PA nocturne et pourrait être proposée, idéalement une fois par an, chez tous les patients. [less ▲]

Jupiter’s dynamic auroral region is the signature of magnetosphere-ionosphere coupling. Precipitating auroral electrons are part of a current system which transports angular momentum from the planetary atmosphere to sub-corotating magnetospheric plasma. The magnitude of the currents and hence precipitating energy flux, are sensitive to the characteristics of the high-latitude magnetosphere, in particular the location of the auroral acceleration region (AAR) and the density and temperature of the high-latitude electron population. We use HST STIS observations of Jupiter’s aurora (Gustin et al. [2016]) to infer the location of the AAR and the properties of the precipitating auroral electrons. To do this, we determine the energy of the precipitating electrons and incident energy flux for the two distinct regions within the main aurora and within flare regions. The resulting relationships between energy flux and electron precipitation energy for the main auroral emission are then compared to the theoretical relationship derived by Lundin & Sandahl [1978], in order to derive the location of the AAR and the temperatures and densities of the electrons at the top of the AAR prior to acceleration. We find that that each emission region is best reproduced using a multiple auroral acceleration regions with different properties, rather than a single auroral acceleration region with a varying potential drop strength. The Hisaki satellite is the first space telescope that is dedicated to observations of our solar system bodies. Hisaki has been continuously monitoring the atmosphere and magnetosphere of the solar system bodies with the extreme ultraviolet (EUV) spectroscope EXCEED since the launch in September 2013. Dynamics on timescales from 10s minutes to a few years were discovered in the atmosphere and magnetosphere by the continuous monitoring. Large joint observing campaigns for Jupiter by the Juno spacecraft in coordination with Hisaki, the Hubble Space Telescope (HST), and other facilities started in mid-2016. Before Jupiter Orbit Insertion, Juno made the in-situ solar wind measurements while Hisaki was simultaneously monitoring Jupiter's aurora and Io plasma torus. On Day of Year 142, Hisaki discovered that the EUV auroral emission power rapidly increased by a factor of ~3 but returned to a nominal level within one planetary rotation. The transient aurora was followed by brightening of the torus sulfur and oxygen ions that is proxy of the hot electron populations in the torus. At the same time, the relatively weak solar wind shock signatures were found in the JUNO/JEDI energetic particle data (Clark et al.). The transient variability is compared to the morphology observed by HST (Nichols et al.). Hisaki is going to monitor Jupiter in collaboration with the large observing program of HST (Grodent et al.) that will make ~150 exposures from this November. [less ▲]

The primary goal of this HST campaign is to complement Juno-UVS (Ultraviolet Spectrograph) observations. This complementarity is four-fold as HST observes Jupiter’s aurora when: 1) Juno-UVS is turned off, that is about 98% of Juno’s 14-day orbit, and Juno’s in situ instruments are in operation. 2) Juno-UVS is operating, but observes the opposite hemisphere of Jupiter. 3) UVS is on in the same hemisphere, but too close to Jupiter to have a global, contextual, view of the aurora and/or UVS is affected by the noise induced by Jupiter’s radiation belts. 4) Juno is too far from Jupiter to get a detailed view of the aurora. In addition, HST will observe the auroral and airglow emissions of the Galilean moons Io, Ganymede and Europa, when UVS is measuring their auroral footprints in Jupiter’s ionosphere. During this campaign, HST is obtaining 45-min STIS time-tag images -movies- of both hemispheres of Jupiter and STIS/COS spectra of Jupiter's moons. These observations are taking place during 4 sequences of Juno's orbit (Figure: typical orbit in magnetic coordinates): 1) Perijove segment: a 6-hour sequence bracketing the time of Juno's closest approach of Jupiter. 2) Crossing segments: few hours periods during which Juno is crossing the magnetic equator of Jupiter and in situ instruments are observing the plasma sheet particles. 3) Perijove +/- 1 Jovian rotation (or more), to provide a context for the auroral activity before and after perijove. 4) Apojove segment: a 12-hour period bracketing the time when Juno is farthest from Jupiter and Juno-UVS is continuously monitoring the global auroral UV power of Jupiter. During Juno orbit PJ5, between 28 Nov. and 07 Dec. 2016, HST obtains 9 STIS movies: 3 movies of the northern aurora near perijove, 1 movie (north) one Jovian rotation before and 2 movies (south- north) one and two Jovian rotations after perijove, 2 movies (north) during two close CS crossings, and 1 movie near apojove. Vertical matter transfer processes (bioturbation and <2μm translocation) in soils and their dynamics are up to now poorly constrained although they are responsible for the temporal evolution of the ecosystem services. In order to better constrain and quantify these processes, a kinetic quantification of the involved transfers based on vertical profiles of 210Pb (xs), 10Be and 137Cs has been developed for Luvisols in which soluble transfers could be neglected. The range of applicability of this type of approach was nevertheless not tested. In this work, we analyzed 210Pb (xs), 10Be and 137Cs in soils exhibiting gradients of organic matter, pH and < 2 μm fraction (Podzol, Andosol Ferralsol, Leptosol). Acidic soils and large organic matter contents were considered. The depth distribution of the studied isotopes was then interpreted as a function of the organic matter content, the soil pH and its < 2 μm fraction. This demonstrates that 10Be and 137Cs losses occur under soluble form in the Podzol, especially under forest, more acid, for the latter. Soluble 10Be transfers were also identified in the Andosol when they were considered as negligible in the Leptosol. For 210Pb (xs), soluble transfers are also probable for soils with pH less than 5.5 (Andosol and Ferralsol) and strong affinity for organic matter has been demonstrated. Thus, under acidic conditions and low < 2 μm fraction, soluble transfer cannot neglected anymore. [less ▲]

Sprinting, jumping and change of direction involve the stretch-shorten cycle (SSC) and therefore plyometric training is used to improve these factors. It has been demonstrated that the jumping strategy utilized can influence the biomechanical profile of the exercise and subsequent adaptation. For example, in some cases, it is important to jump as high as possible while in other cases it is important to reduce the ground contact time. Our hypothesis is that the choice of plyometric exercise has to be matched to the desired biomechanical adaptation. Hence we investigated the influence of 8 weeks of different programs on the plyometric profile of recreational athletes. In each of five groups, very specific exercises were selected according to the training objective: ground contact time group (CT, n=9); vertical jump height group (JH, n=9); CT and JH combination group (CT-JH, n=11); JH + strength training group (JH-S, n=9); and control group (CO, n=8). The plyometric profile performed prior to and post training included measures of jump height, contact time, stiffness and reactivity at different bounding intensities. The results demonstrated that JH and JH-S programs were more effective for improving jump height performance (+7-9%; p<0.005) compared to insignificant jump height changes in the CT and CO groups. CT-JH was the most effective on the reactivity index (+14%, p<0.005) although significant increases (+8%, p<0.05) were also observed in CT, JH and JH-S groups. CT was the only group to significantly decrease short contact time (-5%, p<0.05). ANOVA analysis revealed significant groups*session effect for jump height (p<0.01) and reactivity (p<0.005) but not for short contact time. The present study confirms that the principle of specificity is fundamental in plyometric training so the exercise selection should be developed cognizant of intended adaptations. Jump height and reactivity appear to be differentially affected by specific training practices. [less ▲]

Gammaherpesviruses are important pathogens in human and veterinary medicine. During co-evolution with their hosts, they developed many strategies allowing them to shed infectious particles in presence of immune response. Understanding these strategies is likely to be important to control infection. Interestingly, we recently observed that Murid herpesvirus 4 (MuHV-4), a gammaherpesvirus infecting laboratory mice, could be sexually transmitted between mice. This model offers therefore the opportunity to understand the mechanisms underlying natural transmission. Some of these mechanisms could rely on the glycoprotein 150 (gp150), which could limit virus neutralization and promote the release of infectious particles from cells. In this study, we tested therefore the importance of gp150 in the context of MuHV-4 sexual transmission. Briefly, female mice were infected with WT or gp150- strains expressing luciferase. They were imaged with an in vivo imaging system to follow infection. When lytic replication was observed in the genital tract, infected females were mated with naïve males to compare the capacity of transmission of the two strains. Our results show that, while the gp150- strain has no deficit in reaching and replicating in the female genital tract, it displays a major deficit of sexual transmission in comparison with WT virions. Interestingly, this deficit appears to reflect a deficit of virions release from vaginal epithelial cells. Altogether, our results show that, while gp150 is not required for efficient dissemination and maintenance of MuHV-4 within its host, it is essential for efficient transmission, by promoting the releasing of infectious particles from the mucosal cells. A wide variety of techniques have been developed to determine surface tension, but those that give dynamic measurements, rather than equilibrium measurements of surface tension, are more suitable to study functional properties. Furthermore, the surface activity of surfactants depends on the mechanical properties of the films that are formed at the interface, which are strongly influenced by the intramolecular interactions established. INTRODUCTION : L’hétérogénéité de la dépression est de plus en plus mise en évidence en termes de profils de symptômes, de réponse au traitement, etc. Les théories comportementales de la dépression soulignent l’importance de l’inactivité ainsi que des faibles niveaux de plaisir et de fréquence d’activités plaisantes dans le maintien de la dépression. Cette étude poursuit deux objectifs : (1) investiguer l’hétérogénéité de la depression via une analyse en clusters, (2) investiguer les valeurs prédictives de l’inactivité ainsi que des faibles niveaux de plaisir et de fréquence d’activités plaisantes sur la dépression en fonction du cluster d’appartenance. METHODE : Des adultes tout venant et/ou consultant des centres de santé mentale ont été recrutés (âge entre 18 et 60 ans). Ils ont complété l’Inventaire de Dépression de Beck – Seconde Edition, l’Echelle d’Activation Comportementale pour la Dépression – Version courte, et l’Inventaire des événements plaisants. RESULTATS : Une solution en six clusters a été mise en évidence : “gros dormeurs/heavy sleepers”, “dépressifs cognitifs/cognitive depressives”, “dépressifs affectifs-somatiques/affective-somatic depressives”, “dépressifs légers/mild depressives”, “dépressifs dormant peu/sleepless depressives”, “dépressifs typiques/typical depressives”. Des relations significatives entre inactivité, fréquence d’activités plaisantes et dépression ont seulement été observées chez les “dépressifs cognitifs/cognitive depressives”, les “dépressifs légers/mild depressives” et les “dépressifs typiques/typical depressives”. DISCUSSION : Les résultats confirment l’hétérogénéité de la dépression puisque six clusters ont été mis en évidence. Par ailleurs, l’activation comportementale ne semble pertinente que pour trois de ces clusters dans lesquels les postulats du modèle comportemental de la dépression sont confirmés (“dépressifs cognitifs/cognitive depressives”, “dépressifs légers/mild depressives”, “dépressifs typiques/typical depressives”). La Perspective Temporelle Equilibrée fait référence à une disposition individuelle caractérisée par le fait de se projeter mentalement dans le futur et dans le passé de manière positive et fréquente. L’objectif de notre étude était de valider la version française d’une échelle, initialement conçue en anglais, évaluant cette orientation temporelle : la “Balanced Time Perspective Scale” (Webster, 2011). Une étude en ligne a permis de recueillir les données de 622 participants francophones issus de la population générale. Les résultats indiquent que la version française réplique la structure en deux facteurs du questionnaire original et plaident en faveur d’une bonne validité de construit. De surcroît, les résultats montrent qu’une tendance importante à se projeter mentalement dans le futur et dans le passé de manière positive et fréquente est associée à la réévaluation cognitive, une stratégie de régulation émotionnelle. En somme, ce questionnaire permet d’évaluer la perspective temporelle, qui est ici envisagée en tant qu’importante ressource pour le self, et ce, en étroite relation avec les capacités de régulation émotionnelle. Background: SV2A is the most studied isoform of the Synaptic Vesicle 2 proteins, which are involved in the synaptic vesicle trafficking, being important in normal and pathological process, like the epilepsy (1, 2). [18F]UCB-H was developed like a tool to study the role of this isoform with neuroimaging techniques (3, 4). The objective of this study was to evaluate its specificity to this isoform comparing with the others, through a competition assay in rats with ex-vivo autoradiography and mPET imaging. Methods: Forty male Sprague-Dawley were used in ex-vivo autoradiography experiments (N=20) and in microPET imaging (N=20). Animals were pre-treated 30 minutes before the injection of [18F]UCB-H with a dose IP either of vehicle, Keppra (SV2A ligand), UCB068 (SV2B ligand) or UCB054 (SV2C ligand). Ex-vivo autoradiography was carried out 5 minutes after radiotracer injection while mPET images were acquiring with a dynamic scanner of 1 hour. Data were expressed in Standard Uptake Value and then, the area under the curve was calculated for the total process. Results: In ex-vivo autoradiography, ANOVA of two-ways showed statistical significant differences in brain uptake of [18F]UCB-H among the groups pretreated with Keppra or the ligand for SV2B and the control group. Regarding mPET data, statistical significant differences were found between the group injected with keppra and the rest of groups. Conclusion: Even if a considerable affinity between the ligands UCB068 and UCB054, and the receptor for the isoform SV2A exists, it is only detected during the first 5 minutes (ex-vivo technique), being certainly due to a nonspecific binding. This binding is not strong enough to show a direct competition with the radiotracer during a mPET acquisition. These results allow us to conclude that [18F]UCB-H is a suitable radiotracer for the imaging of the isoform SV2A in vivo, allowing us the clinical study about the molecular base of a disease with a high population impact, like the epilepsy. Approximately 15% of the patients with chronic GVHD develop the sclerodermatous form of the disease characterized by multiple organ fibrosis and loss of skin elasticity. Several studies have shown the potential benefits of imatinib, a tyrosine kinase inhibitor, as a treatment of fibrosis in cGVHD due to its ability to inhibit simultaneously PDGF-R and c-Abl pathways which are both involved in fibrosis mechanisms. This work investigates the possible benefits of imatinib on fibrosis in a murine model of sclerodermatous chronic GVHD (sclcGVHD). Methods: Lethally irradiated Balb/cJ mice (7 Gy TBI) were injected i.v. with 10.106 bone marrow cells and 70.106 splenocytes from B10.D2 donnor mice. Mice were then treated with sterile water or imatinib (150 mg/kg/day) by oral gavage from day +7 to day +52 following transplantation. GVHD severity was assessed three times/week with a scoring system encompassing 5 criteria (mice posture, weight loss, activity, hair loss, skin integrity ; 0-1-2 point(s)/criteria). Results: Our results show that imatinib failed to prevent/improve GVHD with a similar evolution of the GVHD severity with no differences between groups. Histological analyses indicate a significant reduction of the phosphorylation level of the PDGR receptor (p = 0,033) and a trend to a decreased level of phosphorylated c-Abl (p = 0,1854). In vivo cell proliferation assay with CFSE were also performed and showed a reduced proliferation of T cells and subsets (CD4, CD8 and Tregs) in spleen, lymph nodes, bone marrow and blood after imatinib treatment. Finally, FACS analyses performed on days +21 and +35 after transplantation did not show any differences in the absolute T-cell counts. Conclusion: Although we have observed a decreased phosphorylation level of PDGR receptor and less proliferation of T cells and subsets in vivo, imatinib failed to alleviate scl-cGVHD in a murine model of severe scl-cGVHD. Zinc and iron are two essential micronutrients for plants. The homeostasis networks of the two metals are intertwined. Arabidopsis halleri is a zinc- and cadmium-tolerant and zinc-hyperaccumulating species, which also present adaptation of its iron homeostasis(1,4). Transcriptomic studies identified genes which are constitu-tively over-expressed in Arabidopsis halleri compared to Arabidopsis thaliana and which may have a role in metal tolerance or accumulation(2-4). Among them, a candidate gene encodes the FRD3 (FERRIC REDUCTASE DEFECTIVE 3) protein, a member of the MATE family of membrane transporters. FRD3 is a citrate transporter involved in iron homeostasis(5-7) and plays a role in zinc tolerance in A. thaliana(8). The FRD3 gene displays a complex regulation. In A. thaliana, alternative transcript initiation for FRD3 determines two transcripts, which dif-fer in their 5'UTRs and have differential translation efficiency. The two transcripts are selectively regulated under stress conditions: iron and zinc depletion, zinc excess or cadmium presence(9). Natural T regulatory cells (Treg) are challenging to expand ex vivo, and this has severely hindered in vivo evaluation of their therapeutic potential. 5-Azacytidine (5-azaC) and Rapamycin (RAPA) are immunosuppressive drugs that promote selectively the expansion of CD4+CD25highFoxp3+ regulatory T cells. We investigated whether 5-azaC and RAPA could be used together to promote the ex vivo expansion of Tregs purified from adult human peripheral blood. : We found that 5-azaC helped maintain FOXP3 expression during the expansion process probably by promoting the conversion of T conventional (Tconv) in Treg, instead Rapa induces selectively apoptosis in Tconv cells and expansion in Treg. Addition of 5-azaC to RAPA treated cultures improved gene expression of FOXP3, CD25, STAT5 and TGF-B resulted in enhanced Treg expansion and suppressive activity. Also Rapa and 5-AzaC combination sustain Bcl-2 protein expression in Treg conferring resistance to apoptosis process. 5-azaC may have utility in ex vivo expansion of human Tregs, not as a single agent, but in combination with RAPA. These data may considerably accelerate the development of immunotherapeutic approaches for the treatment of autoimmune disease or posttransplant alloreactions by the adoptive transfer of nTreg cells. In Bacillus licheniformis 749/I, the induction of BlaP beta-lactamase relies on a complex regulation system. During this process, the intracellular repressor BlaI is inactivated by a dipeptide coming from the peptidoglycan (PG) degradation via an “AND Gate” regulation. This regulation involves the cellular stress induced by the beta-lactam, the membrane receptor BlaR1 and the PG turnover. Briefly, the induction occurs when the extracellular domain of BlaR1 is acylated by the antibiotic which leads to a reorganization of the transmembrane segments and the receptor autocleavage. Simultaneously, the beta-lactam partially inhibits the penicillin-binding protein 1 (PBP1), triggering increased PG turnover and accumulation of PG fragments. Some of these fragments could enter in the cytoplasm and undergo enzymatic degradations which lead to the formation of the pro co-activator (tripeptide L-Ala-D-Glu-m-A2pm). This pro co-activator generates the co-activator, the dipeptide D-Glu-m-A2pm. Nowadays the nature and the concentration of PG fragments inside the cytoplasm are unknown. Therefore, the development and the validation of a zwitterionic hydrophilic interaction liquid chromatography coupled to electrospray ionization mass spectrometry method (ZIC-HILIC-MS) are required in order to identify and quantify those cytoplasmic fragments. Antioxidants are enzymes, non-enzymatic proteins, minerals micronutrients, vitamins, phenolic compounds, pigments and others. Polyphenols have the ability to reduce or prevent damage caused by free radicals promoting positive effects on human health due to their antioxidant capacity. In this study we evaluated the content of polyphenols from several commercial biological (n = 6) and conventional juices (n = 6 ) (orange, blackcurrant, apple, grapes, tomato, carrot, pomegranate, lemon, grapefruit juices) and we determined the antioxidant effect by inhibition of the superoxide anion production measured by chemiluminescence technology. Conventional juices (orange, apple, tomato, pomegranate, lemon, grapefruit juices) showed more total polyphenolics (+ 32%), more flavan-3-ols (+ 67%) and more flavonols (+ 42%) than the same biological juices. The presence of anthocyanins was reported in juices made with blackcurrant, pomegranate and grapes only, with values above 50 µg/mL, regardless of their nature. Surprisingly, chemiluminescence results showed an inhibition in superoxide anion production by 51% for biological juices vs 23% in conventional ones. The correlation between total polyphenol content and antioxidant capacity of all the juices was r = 0.83 (r2 = 0.7). The fact that there is less polyphenols and higher antioxidant activity in bio juices could be related to the concentration and the type of polyphenol present. [less ▲]

In the Polar Regions, Cyanobacteria are the key primary producers and main drivers of the food webs in a wide range of aquatic to terrestrial habitats. For example, they build benthic microbial mats in lakes and soil crusts. Their success in these harsh cold conditions can probably be explained by particular adaptations to survive freeze/thaw cycles, seasonally contrasted light intensities, high UV radiations, dessication and other environmental stresses. The BCCM/ULC public collection is funded by the Belgian Science Policy Office since 2011. It has obtained the ISO9001 certification for deposition and distribution of strains, as part of the multi-site certification for the BCCM consortium. This collection aims to gather a representative portion of the polar cyanobacterial diversity with different ecological origins (limnetic mats, soil crusts, cryoconites, endoliths,….) and make it available for researchers to study the taxonomy, evolution, adaptations to harsh environmental conditions, pigments, and genomic make-up. It presently includes 226 cyanobacterial strains, of which 119 are of Antarctic origin (catalogue: http://bccm.belspo.be/catalogues/ulc-catalogue-search). As shown by morphological identification, the strains belong to five orders (Synechococcales, Oscillatoriales, Pleurocapsales, Chroococcidiopsidales and Nostocales). The 16S rRNA and ITS sequences of the strains are being characterized. The first 85 Antarctic strains already studied are distributed into 25 Operational Taxonomic Units (OTUs = groups of sequences with > 97,5% 16S rRNA similarity), and thus, represent a quite large diversity. Moreover, strains identified as members of the genera Leptolyngbya or Phormidium appear in several lineages. This supports the idea that there is a need to revise the taxonomy of these polyphyletic genera with a simple filamentous morphology. To better understand the functioning, metabolism and adaptative strategies of cyanobacteria in the extreme Antarctic environment, the genome sequencing of 11 strains has been started. Pair-read data from illumina MiSeq runs were obtained and submitted to a bioinformatic pipeline dedicated to the assembly of genomes and search of sequences involved in the biosynthesis of secondary metabolites. Gene cluster prediction analysis allowed to characterize 20 clusters of NRPS, PKS and hybrid NRPS-PKS from 2 to 66kb. Surprisingly, none of the characterized operons had previously been described in the literature. [less ▲]

In the actual context, biopesticides have emerged as a main alternative to conventional agriculture1. Hence, elicitors are metabolites naturally produced by microorganisms, pathogenic or not, and plants which are able to induce the natural resistance of plants. Also, they have proved to be excellent candidates for biological control. In this context, the lipoxygenase pathway leads to the formation of fatty acid degradation products, called oxylipins, which appear to be crucial agents in plant defence mechanisms2,3. Moreover, with their broad spectrum of action and their possible inducibility, oxylipins appear to be promising candidates for their use as elicitors4. This work focuses on two hydroperoxy-derived oxylipins, the 13(S)-hydroperoxy-octadecadienoic acid (13-HPOD) and the 13(S)-hydroperoxy-octadecatrienoic acid (13-HPOT). The study of the interaction of such compounds with representative plant plasma membrane lipids is essential to understand plant resistance mechanisms. Several in silico and experimental techniques of biophysics showed that acyl-hydroperoxides have significant adsorption capacity and a strong affinity for model membranes. They may also penetrate biological membrane but no permeabilisation effect was observed in this work. Slight conformational differences seem to have a significant impact on their ability to interact with plant plasma membranes. Based on these results, further investigation of the interactions of fatty acids hydroperoxides, even more on the 9-forms, with plant plasma membranes and eventually in the presence of phytopathogenic species, would allow a better understanding of the innate immunity and, on the longer term, could lead to the development of new elicitors with biological mechanisms potentially independent of membrane protein receptors. Supercritical fluid chromatography (SFC) is an attractive separation method often showing higher resolution and shorter analysis time compared to traditional GC and HPLC [2]. Few papers have reported on the SFC analysis of saccharides using standard compounds [3-4]. The aim of this study was to develop an efficient and sensitive method for the determination of fructose, glucose and sucrose in honey samples using SFC-MS. Die Versuche zeigen, dass EDF für verschiedenste Arten von Elektroschrott eingesetzt werden kann. Verschweisste Kompaktgeräte können zum Beispiel so weit geöffnet werden, dass die ge-wünschte Komponente freigelegt und entfernt werden kann. Leiterplatten haben die Eigenschaft, dass bei kontinuierlich steigendem Energieeintrag zuerst die aufgebrachten Komponenten abgetrennt werden, bevor die Cu-Bahnen freigelegt werden. Bei noch höherem Energieeintrag wird die Leiterplatte schlussendlich zerkleinert und vollständig zer-stört. Durch diese Entfrachtung der Leiterplatte kann ein metallreiches Konzentrat hergestellt werden. [less ▲]

Bulk High-Temperature Superconductors (HTS) can be used as efficient passive magnetic shields with performances exceeding those of conventional ferromagnetic materials. The most common geometry for a superconducting shield is a bulk hollow cylinder. In this work, we investigate the magnetic shielding properties when the tube is not in the virgin state. Such a situation happens in practice when the tube has been initially subjected to a first magnetic field. The induced superconducting currents that are flowing in the tube generate a trapped magnetic moment which may deteriorate the shielding of a second field. Here we study experimentally how an initial trapped moment affects the shielding of a magnetic field, when they are both perpendicular to each other. In particular, we focus on the effect of an axial trapped moment on the transverse shielding performances and on the effect of a transverse trapped field on the axial or the transverse shielding performances. Finally, we show how the pristine state of the tube can be restored when applying an external field of adequate amplitude. The consequence for practical applications is that it is possible to recover shielding properties similar to those obtained in the virgin state without heating up the shield above its critical temperature TC. In the frame of the Brain FaCE-It project (Functional biodiversity in a Changing sedimentary Environment: Implications for biogeochemistry and food webs in a managerial setting), the impact of fining and hardening resulting from dredging and wind farms installation on the sediment grain size distribution has to be assessed at the scale of the Southern Bight of the North Sea (SBNS) with a particular focus on the Belgian Coastal Zone (BCZ). With this aim, the ROMS-COAWST tri-dimensional (3D) hydrodynamic model is implemented to simulate the hydrodynamics in the SBNS. At its open boundaries with the Atlantic Ocean and the North Sea, the model is forced with the results of a coarse resolution model available from Mercator. A high resolution of 250 m is used in the area of the BCZ where the accuracy of model predictions needs to be refined. Model currents, tides, temperature and salinity fields will be described and first validation exercises with satellite and local data will be presented and discussed in regards with the objectives of FaCE-It. In a next step, the model will be coupled with a sediment transport in order to describe the dynamics of suspended particulate materials (SPM) and the distribution of the seafloor sediment grain size. When finalized the hydrodynamic model will be coupled with a diagenetic model and will provide environmental conditions for scaling up local foodweb studies that are performed in the frame of FaCE-iT. The final aim is to assess the impact of substrate modifications due to aggregate extraction and wind farms on the biogeochemistry, benthic functionality and food webs at local (around the wind farm) and regional scales (SBNS). Deletion of the growth hormone-releasing hormone gene (Ghrh) results in a severe deficiency of the somatotrope GHRH-GH-IGF-1 axis causing dwarf phenotype that can be reversed by GH or GHRH supplementation (Alba & Salvatori, Endocrinology 2004;145:4134). In basal conditions, the immunological phenotype of Ghrh-/- mice is not markedly disturbed except for a decrease in B cells and an increase in generation of thymic (t) Treg cells (submitted for publication). These data prompted us to investigate immune responses of Ghrh-/- mice using vaccination and infection by S. pneumoniae as models since the response to both stimuli rely on the innate immune system and B cells. Ghrh-/- mice were unable to trigger production of specific IgM and IgG against serotype 1 pneumococcal polysaccharide (PPS) after vaccination with either native PPS (Pnx23) or protein-PPS conjugate (Prev-13) vaccines. A short GH supplementation to Ghrh-/- mice (1 daily injection of 1 mg/kg GH for 4 weeks) restored IgM and IgG response to Pnx23 vaccine but not to Prev-13. This suggests that GH differently impacts on B-1, marginal zone B-2 or innate B-1 B cells. Furthermore, after intranasal instillation of a non-lethal dose of serotype 1 S. pneumoniae, Ghrh-/- mice exhibited a dramatic susceptibility reflected by bacteremia 24h after infection and a survival limit of 72 h, compared to WT C57BL/6 mice that need only 24h to clear infection. The possible impact of GH deficiency on components of the innate immune system that play an important role in defense of the respiratory tract against pneumococcal infection is under current investigation. Creation of the Q-Codes online knowledge base The list of themes, called Q-Codes has been elaborated in a bottom-up approach, by retrieving, in a qualitative analysis study, the themes most frequently addressed by GPs when they meet in congresses. A taxonomy has been build. A multilingual terminological record has been developed for each of the 182 items identified. This work is freely available online on www.hetop.eu/Q (free access after inscription) and is supported by the Rouen (France) biomedical informatics department of the University of Rouen (France). All the data are available in the Web Ontology language (OWL-2), an ontology-ready format fit for future use in the semantic web. Beside its long-term control by steroids, male sexual behavior is also modulated by neuroestrogens in a dynamic way (within minutes) in a number of species ranging from fishes to mammals. Studies in male Japanese quail have also identified following exposure to a receptive female a rapid decrease in the activity of brain aromatase (AA) the enzyme responsible for the conversion of androgens into estrogens. These effects occur mainly within the medial preoptic nucleus (POM), a sexually dimorphic structure of the preoptic area that plays a key role in the activation of male sexual behavior and contains the highest AA in the brain. In vitro studies demonstrated that AA can be rapidly inhibited by calcium-dependent phosphorylations of the enzyme triggered by the activation of AMPA and kainate receptors. We confirmed here this rapid effect of glutamate on AA by injecting kainate in the POM of anesthetized males and measuring AA in the tissue after brain collection. AA in POM was inhibited in the kainate-injected hemisphere compared to the control hemisphere injected with vehicle. In a second experiment, we showed by in vivo microdialysis that glutamate is released in POM during copulation. These results thus suggest that glutamate controls dynamic changes of AA that occur in the brain during sexual interactions. To confirm that the decrease in AA leads to an actual reduction of local estradiol concentration, we quantified via microdialysis and radioimmunoassay changes in estradiol concentration in the male POM during sexual interactions with a female. Surprisingly, a dramatic elevation of estradiol was observed during copulation. Estradiol has been shown to enhance acutely male sexual motivation, therefore the function of its increase in the POM could be to maintain motivation during the entire sexual encounter. The decrease of AA observed ex vivo after copulation would then reflect a compensatory mechanism to restore baseline pre-copulatory conditions. Importantly, these results highlight that although long-term changes in AA are often used as a proxy for local estradiol concentrations, these two measures can show major short-term discrepancies possibly reflecting variations in estrogen turnover. Spatial autocorrelation expresses the dependence between values at neighbouring locations. Testing if the spatial autocorrelation is significant may confirm or deny the need to consider spatial analysis over the classical one. Indeed, the analysis of spatial data is only meaningful if the spatial components bring information. Several measures of spatial autocorrelation are defined in the literature. Moran’s index, Geary’s ratio and Getis-Ord statistic are the most used statistics. Tests based on these measures have been developed in the literature using asymptotic and permutation results. They are used in practice in many fields, for instance in geography, economics, biogeosciences, medicine, ... However, these tests should be cautiously applied because they are not robust. A single contaminated observation can significantly modify their results. This poster is composed of two parts. Firstly, the robustness of already available tools for measuring spatial autocorrelation will be analysed. Secondly, alternative methods will be proposed to robustly test the spatial autocorrelation. Introduction: Des stéréotypes négatifs liés à l’avancée en âge sont fréquemment observés en population générale. Des études ont montré que ces stéréotypes sont plus puissants que ceux liés à l’origine ethnique, au sexe ou à l’orientation sexuelle. Lorsqu’ils sont présents chez les soignants, ces stéréotypes peuvent entrainer des discriminations et limiter l’accès aux soins des personnes âgées (Schroyen et al., 2016). Il a en outre été démontré qu’un comportement d’aide injustifiée pouvait favoriser la perte d’autonomie chez une personne âgée. L’objectif de cette étude est d’évaluer la force des stéréotypes âgistes chez les étudiants en médecine et de rechercher les facteurs qui y sont associés. Méthodes: Un questionnaire écrit informatisé a été proposé aux étudiants en médecine des facultés de Rennes, Brest et Liège, de la 2ème à la 6ème année. Il associe trois mesures : l’Adaptation française de Fraboni Scale of Ageism, une tâche de fluence verbale à propos du vieillissement et de la jeunesse, et une mesure de la stigmatisation liée à l’âge par estimation de pourcentages. Résultats: Des données ont été obtenues pour 570 étudiants à Rennes et 485 à Liège; le recueil de données se poursuit à Brest. Les premières analyses confirment que l’âgisme est largement répandu chez les étudiants en médecine, avec des résultats globalement similaires à Rennes et Liège. Les étudiants de sexe masculins, ceux qui ont peu de contact avec des personnes âgées, ou qui ont des contacts jugés de mauvaise qualité, sont plus susceptibles d’exprimer des stéréotypes âgistes. Le fait d’avoir fait un stage dans un service de gériatrie ou d’avoir bénéficié d’un enseignement de la gériatrie ne semble pas influencer significativement ces stéréotypes. Conclusion: L’enseignement de la gériatrie aux étudiants en médecine doit prendre en compte la fréquence des stéréotypes âgistes. Nos résultats étant soumis à un biais de désirabilité sociale, des études portant sur les stéréotypes implicites (inconscients) seraient utiles. Une meilleure connaissance de l'âgisme et de ses déterminants chez les étudiants en médecine est nécessaire à l'élaboration d'approches pédagogiques susceptibles de limiter les conséquences de ces stéréotypes sur la qualité des soins aux personnes âgées [less ▲]

Background The creation of arteriovenous fistula (AVF) may retard chronic kidney disease progression in the general population. Conversely, there is limited literature regarding the impact of AVF closure on renal function in kidney transplant recipients (KTR). Methods All KTR were retrospectively identified from 01/01/2007 to 31/12/2013, and grouped into: (0) no AVF; (1) closed AVF; and (2) left open AVF. Glomerular filtration rate was estimated (eGFR) upon MDRD and FAS equations. Linear mixed models calculated the slope and intercept of eGFR decline versus time, starting at 3 months post transplantation (Tx). Comparative analyses of eGFR slopes were performed among groups, as well as before vs after AVF closure in group 1. For the latter, time was balanced before vs after AVF closure, with at least 10 observations per patient. Results The cohort included 285 KTR (Table 1), with a median follow-up of 1750 days [1665; 2028]. Focusing on group 1, AVF closure occurred after a mean time of 653 ± 441 days post Tx, with a thrombosis/ligation ratio of 19/95. Balanced study periods before vs after AVF closure lasted 15.7 and 14.9 months, respectively. No difference was found between corresponding intercepts (p, 0.11). By contrast, eGFR slopes were significantly different before (0.043 ml/min/year) versus after (-0.176 ml/min/year) AVF closure (p, 0.0115). Similar observations were obtained using FAS equation Conclusion A significant acceleration of eGFR decline is observed over the 15 months following the closure of functioning AVF in KTR. [less ▲]

In Europe, ruminant processed animal proteins (PAPs) and blood products are not allowed to be used in feed for farmed animal. In contrast, blood meal and blood products of porcine origin are both authorised in aqua feed, whereas only porcine blood products are allowed to be used in feed intended for other non-ruminants. Besides official methods (light microscopy and PCR), complementary methods are developed in order to refine the by-products identification. By-products derived from blood are one of these products for which additional information are needed. Indeed, these prohibited materials sometimes cannot be distinguished from those authorised (e.g. milk powder) using official methods. The aim of this work was to develop a fast and easy method to detect blood meal and blood products. This study was based on the detection of hemoglobin in animal feed by synchronous fluorescence spectroscopy (SFS). To achieve this goal, preliminary tests were carried out on reference material (hemoglobin and albumin purchased from Sigma Aldrich) in order to determine appropriate conditions (solvent) and parameters (offset values) for hemoglobin detection. Selected settings were then applied to analyse protein extracts of commercial feed material derived from blood. The results obtained on blood meal and blood products (hemoglobin powder and plasma powder) showed fluorescence spectral bands that characterise hemoglobin. In order to determine whether the method was fit for the purpose, real commercial compound feeds known to contain or to be free from blood products or blood meal were analysed. The Principal Component Analysis (PCA) applied to these spectra showed that it was possible to discriminate samples containing hemoglobin from those that do not contain. This result confirmed that SFS is a promising screening method for the detection of hemoglobin in animal feed. [less ▲]

CXCR7 is an atypical and enigmatic chemokine receptor playing crucial roles in numerous physiological processes but also in viral infection and cancer. To date, the exact functions and signaling properties of CXCR7 are still a matter of debate. Unlike other chemokine receptors, CXCR7 is proposed to act as a scavenging receptor for its ligands, to trigger arrestin-dependent signaling and possibly classical G-protein mediated signaling. Besides its two endogenous chemokines CXCL11 and CXCL12 and the virus-encoded chemokine vCCL2, accumulating evidence suggest that CXCR7 interacts with several non-chemokine ligands such as the pro-angiogenic peptide Adrenomedullin (ADM). Furthermore, calcitonin receptor like receptor (CLR) and receptor activity modifying proteins (RAMPs), constituting the main ADM receptor, might crosstalk with CXCR7 conferring the receptor alternative functions. While the interactions of CXCR7 with classical chemokine ligands are now well characterized, little is known about the interdependence of ADM and the membrane interacting partners with CXCR7, the signaling pathways activated as well as their roles in viral infection and tumorigenesis. The aims of this PhD project are to 1) unravel the role and impact of CLR and RAMPs on the binding, scavenging and signaling properties of CXCR7 towards the different ligands ADM, CXCL12, CXCL11 and vCCL2. Additionally, by using state-of-the-art techniques like Bioluminescence Resonance Energy Transfer (BRET), we will 2) inquire the ability of CXCR7 to heterodimerize with CLR and RAMPs. Lastly, since CXCR7 and RAMPs were described to be upregulated upon infection by several tumorigenic viruses, 3) a detailed expression profile of all the ligands and partners of CXCR7 in a herpesvirus-induced cancer model will be determined and will help to reveal the role of CXCR7 in viral associated tumorigenesis. This project will allow improving our level of understanding of CXCR7 regulation and functions and will open new therapeutic directions for selective CXCR7 inhibition. [less ▲]

The aim of this study was to compare the effect of aging conditions (dry-aging vs. wet-aging) and time (7, 28, 49 and 70 days) on tenderness, color and lipid stability of Charolais beef. Three longissimus dorsi of Charolais cows were dry- or wet-aged for up to 70 days at 2 °C. At different times, part of these samples was cut into steaks, vacuum packaged and stored during 4 days at 4 °C + 8 days at 8 °C. The following parameters were evaluated at different intervals: pH, tenderness (Warner–Bratzler peak shear force), color (CIE L*a*b*), myoglobin oxidation (K/S572:K/S525 ratio) and lipid oxidation (TBARS). No difference in tenderness was observed between the two aging techniques. The sensitivity of samples to oxidation was influenced by the aging technique (wet-aging > dry-aging). For the studied samples, aging beyond 28 days could be considered excessive from an organoleptical point of view. [less ▲]

Blood derived products are valuable animal products used in feed for their nutritional value and their positive effects on growth and health. Nevertheless, since the BSE crisis, their use is strictly regulated. Blood meal and blood products (hemoglobin powder and plasma powder) of bovine origin are totally prohibited. Blood meal and blood products of porcine origin are authorised in aquafeed, whereas only blood products are allowed to be used in feed intended for other non-ruminant. The detection of the type of protein and the species of origin is therefore crucial to ensure feed safety. With the current official methods for the detection of PAPs, light microscopy and PCR, it is not always possible to specifically identify this type of protein source. The objective of our work was to set-up a routine mass spectrometry method for a qualitative detection of blood meal and hemoglobin powder of bovine and porcine origin in feed. The method was based on the detection of species-specific peptides biomarkers identified in a previous study by a non-targeted approach. All biomarkers were peptides of α and β hemoglobin subunits. Proteins were extracted using a TCA/acetone protein precipitation protocol followed by purification with a 2-D Clean-Up Kit (GE Healthcare, USA). After an in-solution trypsin digestion step, analyses were performed by liquid chromatography (Acquity system, Waters, UK) coupled with a triple quadrupole mass spectrometer (Xevo TQS, Waters, UK) with electrospray ionisation. The acquisition and processing of data was carried out by MassLynx software (v. 4.1, Waters) and multiple reaction monitoring (MRM) design was made using the open-source software Skyline (https://skyline.gs.washington.edu/labkey/project/home/software/Skyline/begin.view). Reference hemoglobin powder was used to select MRM transitions for each peptide biomarkers and to optimise their collision energy. Commercial feed material and compound feed containing or free from blood derived products were then analysed. Selected transitions were present in materials or feed known to contained blood derived product of the same origin and absent in the others. Artificially contaminated feed with various contamination levels were also analysed in order to evaluate the influence of matrix composition and to experimentally determine the limit of detection. A first estimation of the LOD was around 0.05 % w/w which was below the LOD imposed by the EC for animal proteins detection method. [less ▲]

In the Polar Regions, Cyanobacteria represent key primary producers and are the main drivers of the food webs in a wide range of aquatic to terrestrial habitats. For example, they build benthic microbial mats in lakes and soil crusts in terrestrial biotopes. They may present interesting features to survive freeze/thaw cycles, seasonally contrasted light intensities, high UV radiations, dessication and other stresses. The BCCM/ULC public collection funded by the Belgian Science Policy Office since 2011 aims to gather a representative portion of the polar cyanobacterial diversity with different ecological origins (limnetic mats, soil crusts, cryoconites, endoliths…). It makes it available for researchers to study the taxonomy, evolution, adaptations to harsh environmental conditions, and genomic make-up. It presently includes 226 cyanobacterial strains, with 119 being of Antarctic origin (catalogue: http://bccm.belspo.be/catalogues/ulc-catalogue-search). An ISO 9001 certificate was obtained for the public deposition and distribution of strains, as part of the multi-site certification for the BCCM consortium. The morphological identification shows that the strains belong to the orders Synechococcales, Oscillatoriales, Pleurocapsales, Chroococcidiopsidales and Nostocales. The 16S rRNA and ITS sequences of the strains are being characterized. Our results show that the Antarctic strains are positioned into 25 OTUs (sequences with > 97,5% 16S rRNA similarity), and thus, represent a quite large diversity. In addition, cyanobacteria are known to produce a wide range of secondary metabolites (e.g. alkaloids, cyclic and linear peptides, polyketides) with bioactive potential. Among these bioactive metabolites, some display antibiotic, anticancer or antifungal effects. In collaboration with the BCCM/IHEM collection of biomedical fungi, a screening of cyanobacterial strains from BCCM/ULC was performed in order to discover potential new antifungal drugs. The analysis of a first set of methanol extracts from 15 different strains put in evidence the antifungal activity of a Phormidium priestleyi isolate. The latter remains active up to 0.5% (v/v) of fungal culture and was able to inhibit the growth of various fungal species among Candida, Cryptococcus, Aspergillus, and Penicillium. The raw extract was subjected to HPLC and a fraction containing the active molecule was obtained. This molecule appeared to be a thermostable hydrophobic compound. Moreover, in vitro toxicological analyses suggest that the compound has a general cytotoxic effect that could be inhibited by the mammalian metabolism. Further analyses are needed to identify the molecule and to determine if it could be a candidate for a new antifungal drug. In summary, the BCCM/ULC public collection serves as a Biological Resource Centre to conserve ex situ and document the biodiversity of polar cyanobacteria, as well as a repository for discovery of novel bioactive compounds. Over the last decades, the Arctic has experienced a warming trend that is nearly twice as high as the global average, a phenomenon known as ‘Arctic amplification’. The impact of warmer temperatures on Arctic ecosystems is still unclear. Cyanobacteria are the key primary producers in freshwater and terrestrial Arctic ecosystems, where they are the driver for numerous ecological functions. For a better understanding of the impacts of climate change on Arctic ecosystems, baseline knowledge on cyanobacterial diversity and distribution is crucial. Here we investigate, for the first time, the biogeographic patterns of cyanobacterial communities across Svalbard, using 454 pyrosequencing of partial 16S rRNA gene sequences. Samples were taken from distinct ecosystems and biogeographic zones. We also compare the studied communities with similar Antarctic communities. [less ▲]

Les pucerons font partie des insectes qui causent des dégâts d’importances économiques sur une grande diversité de plantes cultivées. Les insecticides de synthèse généralement utilisés pour les contrôler montrent de plus en plus leurs limites (résistance), et sont néfastes pour l’homme et l’environnement. Dans la recherche de méthodes de lutte alternatives, le comportement de choix d’Acyrthosiphon pisum Harris (Homoptera : Aphididae) exposé à des sources d’odeurs de plant de fèves (Vicia fabae L.) et du basilic européen (Ocimum basilicum L.) à l’aide d’un olfactomètre en Y a été étudié. Les observations de cette expérience montrent que les pucerons se dirigent préférentiellement vers la source d’odeurs de fève et y passent plus de temps. Ce résultat montre que les volatiles du basilic ont un effet répulsif sur les pucerons du pois. D’autres tests seront évalués sur des espèces de pucerons plutôt généralistes afin de valider l’effet répulsif du basilic sur les pucerons. [less ▲]

Halyomorpha halys Stål, la punaise diabolique, est une espèce invasive préoccupante en provenance d’Asie de l’Est. En moins de 15 années, elle a colonisé près de l’ensemble des Etats-Unis d’Amérique, occasionnant des pertes de rendement considérables dans les vergers et les cultures en champs ainsi que des nuisances dans les habitations durant l’hiver. La punaise diabolique a été recensée en Suisse en 2007, et il est à craindre qu’elle colonise la presque totalité de l’Europe d’ici 2020. Cette étude a pour objectif de prévenir l’installation de la punaise diabolique dans les pays européens en se focalisant sur le contrôle biologique. Des scientifiques américains ont développé une stratégie « attirer et tuer » basée sur la phéromone d’agrégation de la punaise diabolique et des pesticides. D’autres ont montré l’efficacité d’un champignon pathogène d’insectes (Beauveria bassiana strain GHA. Afin d’établir un contrôle rapide du ravageur en Europe, compatible avec l’agriculture biologique, une idée serait de combiner l’effet attractif de la phéromone d’agrégation et l’application de spores de B. bassiana.
