The metallo-β-lactamase (MBL) GOB-1 was expressed via a T7 expression system in Escherichia coli BL21(DE3). The MBL was purified to homogeneity and shown to exhibit a broad substrate profile, hydrolyzing all the tested β-lactam compounds efficiently. The GOB enzymes are unique among MBLs due to the presence of a glutamine residue at position 116, a zinc-binding residue in all known class B1 and B3 MBL structures. Here we produced and studied the Q116A, Q116N and Q116H mutants. The substrate profiles were similar for each mutant, but with significantly reduced activity compared with that of the wild-type. In contrast to the Q116H enzyme, which bound two zinc ions just like the wild-type, only one zinc ion is present in Q116A and Q116N. These results suggest that the Q116 residue plays a role in the binding of the zinc ion in the QHH site. [less ▲]

Geometrical acoustics methods have already been transformed to account for diffusely reflecting boundaries. In randomized ray-tracing algorithms, the sound rays are either specularly reflected or scattered, according to the value of a random number which is compared with the diffusion factor. However, this method becomes inefficient if this factor depends on frequency, since the process must then be repeated for each frequency band. A method is proposed in this paper to compute all frequency components simultaneously in a single pass. The diffuse reflection model is based on the definition of a new concept: the "splitting coefficient," which can differ from the diffusion factor. First, the randomized ray-tracing method is described and the diffusion model is applied to a single diffusing surface in free field. It is shown that the results computed by the program are in accordance with theoretical results. Then, the method is shown to work properly when it is applied to more realistic enclosures: This is proved in theory and illustrated by some examples. A particular problem is the increase of the statistical error which has been solved by an appropriate control of the splitting coefficient. [less ▲]

We examine non-linear resonant interactions between a damped and forced dispersive linear finite rod and a lightweight essentially non-linear end attachment. We show that these interactions may lead to passive, broadband and on-way targeted energy flow from the rod to the attachment, which acts, in essence, as non-linear energy sink (NES). The transient dynamics of this s stem subject to shock excitation is examined numerically using a finite element (FE) formulation. Parametric studies are performed to examine the regions in parameter space where optimal (maximal) efficiency of targeted energy pumping from the rod to the NES occurs. Signal processing of the transient time series is then performed. employing energy transfer and/or exchange measures, wavelet transforms, empirical mode decomposition and Hilbert transforms. By computing intrinsic mode functions (IMFs) of the transient responses of the NES and the edge of the rod, and examining resonance captures that occur between them, we are able to identify the non-linear resonance mechanisms that govern the (strong or weak) one-way energy transfers from the rod to the NES. The present study demonstrates the efficacy of using local lightweight non-linear attachments (NESs) as passive broadband energy absorbers of unwanted disturbances in continuous elastic structures, and investigates the dynamical mechanisms that govern the resonance interactions influencing this passive non-linear energy absorption. Damselfish are prolific callers, identified as being able to produce different kinds of sounds (pops and chirps) associated with various behaviors. During courtship and chase behaviors, the coral reef damselfish Pomacentrus amboinensis produces a previously unknown additional kind of call that we have named the wiping sound. These calls have two to three long (54 ± 13 ms) units that do not show the usual percussive aspect of the pomacentrid sounds. Calls consist of a high-pitched (from 550 to 775 Hz) tonal sound in which the cycle repetition rate corresponds to the peak frequency. The high frequency of this tonal sound can be excluded as coming from the contraction of sound producing muscles. These sounds could be the result of a mechanism that drives the merging of successive pops. In the noisy environment of coral reefs and the resulting competition for acoustic space, the wiping sounds appear to be a good way to increase signal distinctiveness and opportunities for correct signal discrimination. This new kind of sound supports that acoustic communication is highly important in the biology of damselfishes and their diversification. [less ▲]

This article examines how science, technology, and innovation (STI) policies in Flanders (Belgium) are affected by, and potentially transformed through, technology assessments (TAs). Broadly defined, TAs encompass activities and programs that expand and deepen the knowledge base of contemporary knowledge-based economies (KBEs), typically by including new actors (e.g. trade unions), ideas (e.g. science in society), and rationales (e.g. participatory techniques) in STI processes. Starting from the regionalization of STI policy in Belgium and the convergence of Flemish STI around global KBE principles, the article exemplifies how since the 1980s successive Flemish TA waves (early- warning, bottom-up, and interactive TA) have co-evolved with successive generations of Flemish innovation policy. Building on these findings, it argues that Flemish TA has assumed the role of mediator between science and society, both by counteracting and accommodating dominant STI paradigms. By providing a historical and socio-political perspective on TA and innovation policy, the article draws critical attention to the institutional settings and societal contexts in which TA is embedded, and questions TA's strategic utility within contemporary KBEs. This perspective sheds light on the Flemish government's recent decision to close its parliamentary TA institute and the institutional expansion of TA elsewhere in Europe. [less ▲]

1. Due to intensive selection, broiler chickens became the most efficient meat-producing animals because of their fast growth, supported by a virtually unlimited voluntary feed intake. These characteristics cause many problems in the management of broiler breeder hens because of the negative correlation between muscle growth and reproduction effectiveness. 2. This problem, namely the fast muscle growth versus reproduction health paradox, induces a second paradox, acceptable reproduction and health versus hunger stress and impaired welfare, because broiler breeder hens require dedicated programmes of feed restriction (1) to maximise egg and chick production and (2) to avoid metabolic disorders and mortality in broiler breeders. 3. Given that poultry selection is a global large-scale business and chickens are a prolific species, improvement in profit can only be obtained by selecting on feed conversion and/or for higher breast meat percentage, which will intensify the broiler-breeder paradox. 4. New feeding strategies are being studied, but it is questionable if the paradox can be solved by management tools alone. Because breeding and selection are long-term processes, involving animals, farmers, consumers, industry, environment etc., a more sustainable breeding goal needs to be determined by a multidisciplinary approach and an open debate between several actors in the discussion. 5. Using dwarf broiler breeder hens could be one alternative, because dwarf hens combine relatively good reproductive fitness with ad libitum feeding. Another possibility is to accept lower broiler productivity by assigning economic values to welfare and including integrity traits in an extended breeding goal. [less ▲]

Since the end of the civil war in 2009, rural Southern Sri Lanka is facing growing tensions between fast economic development and the conservation of biodiversity, which contributes to the notoriety of the island. In order to preserve the green areas of the country, both the government and international institutions have made great efforts to implement projects combining biodiversity protection and poverty reduction as their objectives. Within that context, this paper focuses on a specific category of stakeholders, emerging from the implementation of the aforementioned projects: a wide range of intermediaries between local communities and the State, or international institutions. Based on an ethnographic study of two protected areas in Southern Sri Lanka, the paper will examine the role of those intermediaries in the implementation of environmental governance models and ideologies in local arenas. I will also analyze the complex relationships between those brokers and other categories of individuals or groups within the community: people living in protected areas, traditional figures of power, social and political elites, etc. Using fieldwork illustrations, I will stress how the emergence of brokers and intermediaries in the context of nature conservation may lead to the reorganization of social and political relationships at the local scale. Very little is known on the exposure of high trophic level species to current-use brominated (BFRs) and phosphorus flame retardants (PFRs), although observations on their persistence, bioaccumulation potential, and toxicity have been made. We investigated the accumulation of BFRs and PFRs, and their associations with dietary proxies (δ13C, δ15N and δ34S), in plasma and feathers of White-tailed Eagle Haliaeetus albicilla nestlings from Trøndelag, Norway. In addition to accumulation of a wide range of polybrominated diphenyl ether (PBDE) congeners in both plasma and feathers, all non-PBDE BFRs and PFRs could be measured in feathers, while in plasma only two of six PFRs, i.e. tris-(2-chloroisopropyl) phosphate (TCIPP) and tris-(2,3-dichloropropyl) phosphate (TDCPP) were detected. PFR concentrations in feathers (0.95-3,000 ng g-1) were much higher than selected organochlorines (OCs), such as polychlorinated biphenyl 153 (CB 153; 2.3-15 ng g-1) and dichlorodiphenyldichloroethylene (p,p’-DDE; 2.3-21 ng g-1), PBDEs (0.03-2.3 ng g-1) and non-PBDE BFRs (0.03-1.5 ng g-1). Non-significant associations of PFR concentrations in feathers with those in plasma (P≥0.74), and their similarity to reported atmospheric PFR concentrations, may suggest atmospheric PFR deposition on feathers. Most OCs and PBDEs, as well as tris(chloroethyl) phosphate (TCEP), tris(phenyl) phosphate (TPHP) and tri-(2-butoxyethyl) phosphate (TBOEP) were associated to δ15N and/or δ13C (all P≤0.02). Besides δ15N enrichment, δ34S was depleted in nestlings from fjords, inherently close to an urbanised centre. As such, both may have been a spatial proxy for anthropogenic disturbance, possible confounding their use as dietary proxy. [less ▲]

BACKGROUND: Immunological and functional bronchopulmonary abnormalities may be present in up to two-thirds of patients with Crohn's disease. Having recently described a mild increase in methacholine airways responsiveness in these patients, we investigated whether this physiological abnormality is associated with bronchial inflammation since it has previously been described in asthma. METHODS: Eighteen patients with Crohn's disease and 15 healthy controls matched for age, atopy and smoking habit, were studied. All the subjects underwent a bronchial methacholine challenge (1, 4 and 16 mg/mL) and a sputum induction by inhalation of hypertonic saline (NaCl 4.5%). The sputum samples were analysed for their cellular composition as well as for the levels of several mediators and proteins in the fluid phase, including eosinophil cationic protein (ECP), myeloperoxydase, albumin, alpha2-macroglobulin, interleukin-8 (IL-8), IgA and IL-8/immunoglobulin A complexes. RESULTS: When compared to control subjects, patients with Crohn's disease had significantly higher sputum eosinophil counts (14.5% [0-79.9%] vs 0.2% [0-2.3%]; P < 0. 001) and ECP levels (26.2 microg/L [4-124.2 microg/L] vs 9.8 microg/L [0-94.2 microg/L]; P < 0.05). However, patients with Crohn's disease had no sign of increased plasma exudation as reflected by sputum levels of albumin and alpha2-macroglobulin similar to those seen in control subjects. Furthermore the sputum levels of IL-8, IgA and IL-8/IgA complexes were not significantly different between the two groups. The magnitude of the fall in forced expiratory volume in 1 s after methacholine inhalation was significantly increased in Crohn's disease patients although it did not correlate with the extent of sputum eosinophilia or with the sputum ECP levels. CONCLUSIONS: Crohn's disease patients without any clinical respiratory involvement have airway eosinophilia without local increased plasma exudation. However, bronchial eosinophilia in Crohn's disease per se is not sufficient to induce clinically significant airway hyperresponsiveness, suggesting that other factors than bronchial eosinophilic infiltration are required for the clinical expression of an airway instability.
