The classical fate of metastasizing breast cancer cells is to seed and form secondary colonies in bones. The molecules closely associated with these processes are predominantly present at the cell surface and in the extracellular space, establishing the first contacts with the target tissue. In this study, we had the rare opportunity to analyze a bone metastatic lesion and its corresponding breast primary tumor obtained simultaneously from the same patient. Using mass spectrometry, we undertook a proteomic study on cell surface and extracellular protein-enriched material. We provide a repertoire of significantly modulated proteins, some with yet unknown roles in the bone metastatic process as well as proteins notably involved in cancer cell invasiveness and in bone metabolism. The comparison of these clinical data with those previously obtained using a human osteotropic breast cancer cell line highlighted an overlapping group of proteins. Certain differentially expressed proteins are validated in the present study using immunohistochemistry on a retrospective collection of breast tumors and matched bone metastases. Our exclusive set of selected proteins supports the set-up of further investigations on both clinical samples and experimental bone metastasis models that will help to reveal the finely coordinated expression of proteins that favor the development of metastases in the bone microenvironment. Aphids are some of the most serious pests of cultivated crops worldwide, causing major yield and economic losses. Previous works have demonstrated ants and natural enemies (including ladybeetles and hoverflies) to be able to use aphid volatile chemicals to locate aphid colonies. Here, we report the first isolation of a bacterium from the pea aphid Acyrthosiphon pisum honeydew, Staphylococcus sciuri, which produces kairomones used by the aphidophagous hoverfly Episyrphus balteatus and the Asian Ladybeetle Harmonia axyridis during their search for prey colonies. Some specific semiochemicals produced by S. sciuri were identified as attractants and ovipositional stimulants. Similarly, we have shown scouts of the aphid tending ant species, Lasius niger, to orientate their foraging behaviour toward an Aphis fabae infested plant and we have demonstrated that the odours released by this aphid honeydew were attractive for ant scouts. Again, bacteria were involved in the production of these honeydew semiochemicals. Interestingly, ant scouts were also able to discriminate honeydew odour from A. fabae (usually attended by L. niger) and A. pisum (unattendedby L. niger). Comparison of the volatile and bacteria composition of both aphid species honeydew were attended. [less ▲]

Analytical method validation is a mandatory step to evaluate the ability of developed methods to provide accurate results for their routine application in order to trust the critical decisions that will be made with them. Even if several guidelines exist to help perform analytical method validations (ICH Q2R1 [1], USP <1225> [2], …) there is still the need to clarify the meaning and interpretation of analytical method validation criteria and methodology. Indeed, actually method validation is mostly realised as the traditional check list implementation of e.g. the ICH Q2R1 or USP <1225> method validation requirements. However, within the trend of Quality by Design [3], there is the need to switch from this traditional vision to an analytical method validation really adding value and providing a high level of assurance of analytical methods results reliability. Yet, different interpretations can be made of the validation guidelines as well as for the definitions of the validation criteria. This will lead to diverse experimental designs implemented to try fulfilling these criteria. Finally, different decision methodologies can also be interpreted from these guidelines. Therefore, the risk that a validated analytical method may be unfit for its future purpose will depend on a personal interpretation of these guidelines. The objective of this presentation is thus to show that analytical method validation should be planned and performed by first starting with the end in mind: what is the objective of the analytical methods under study? In such a way analytical method validation is coherent with the actual Quality by Design regulatory expectations. The risk of having validated an analytical method unfit for its purpose is strongly reduced as well as the risk of generating Out of Specification (OOS) results due to an unfit method. Eddy current problems are addressed in a bidimensional setting where the conducting medium is non-magnetic and has a corner singularity. For any fixed parameter linked to the skin depth for a plane interface, we show that the flux density is bounded near the corner unlike the perfect conducting case. Then as goes to zero, the first two terms of a multiscale expansion of the magnetic potential are introduced to tackle the magneto-harmonic problem. The heuristics of the method are given and numerical computations illustrate the obtained accuracy. [less ▲]

The additional energy spread due to sample porosity was implemented in the SIMNRA simulation code, version 6.60 and higher. Deviations of the path length and energy loss distributions from the ones expected from a Poisson distribution of the number of traversed pores are taken into account. These deviations are due to the interaction of pores at higher pore concentrations by overlap or blocking. The skewnesses of the energy distributions are approximated by two-piece normal distributions with identical first three moments. Propagation of porosity-induced energy spread in thick layers is taken into account. Calculated results are compared to experimental data obtained with thin TiO2 mesoporous films measured by Rutherford backscattering (RBS),transmission electron microscopy (TEM), and atmospheric poroellipsometry. Model parameterization through adjustment to field data is a crucial step in the modeling and the understanding of the drainage network response to tectonic or climatic perturbations. Using as a test case a data set of 18 knickpoints that materialize the migration of a 0.7-Ma-old erosion wave in the Ourthe catchment of northern Ardennes (western Europe), we explore the impact of various data fitting on the calibration of the stream power model of river incision, from which a simple knickpoint celerity equation is derived. Our results show that statistical least squares adjustments (or misfit functions) based either on the streamwise distances between observed and modeled knickpoint positions at time t or on differences between observed and modeled time at the actual knickpoint locations yield significantly different values for the m and K parameters of the model. As there is no physical reason to prefer one of these approaches, an intermediate least-rectangles adjustment might at first glance appear as the best compromise. However, the statistics of the analysis of 200 sets of synthetic knickpoints generated in the Ourthe catchment indicate that the timebased adjustment is the most capable of getting close to the true parameter values. Moreover, this fitting method leads in all cases to an m value lower than that obtained from the classical distance adjustment (for example, 0.75 against 0.86 for the real case of the Ourthe catchment), corresponding to an increase in the non-linear character of the dependence of knickpoint celerity on discharge [less ▲]

Improvements on the Liége cyclotron have yielded an energy resolution comparable to that of classic electrostatic accelerators. The latest upgrades of the High-Energy High-Resolution beam line (HE-HR) are presented which consist of the addition of both a new extraction nozzle for non-invasive in-air measurements and a new particle-detection setup developed to take full advantage of the increased probed thickness with depth sensitive analytical methods. The suitability of the new setup and the advantages provided by the larger flexibility in choosing both the proper particle with an extended energy available will be illustrated by discussing latest results of studies recently led on Roman gilding techniques on cultural heritage artifacts by means of “High-Energy” alpha backscattering spectrometry combined with PIXE. Analytical methods capability evaluation can be a useful methodology to assess the fitness of purpose of these methods for their future routine application. However, care on how to compute the capability indices has to be made. Indeed, the commonly used formulas to compute capability indices such as Cpk, will highly overestimate the true capability of the methods. Especially during methods validation or transfer, there are only few experiments performed and, using in these situations the commonly applied capability indices to declare a method as valid or as transferable to a receiving laboratory will conduct to inadequate decisions. In this work, an improved capability index, namely Cpk-tol and the corresponding estimator of proportion of non conforming results ( ) is proposed. Through Monte-Carlo simulations, they have been shown to greatly increase the estimation of analytical methods capability in particular in low sample size situations as encountered during methods validation or transfer. Additionally, the usefulness of this capability index is illustrated through several case studies covering applications commonly encountered in the pharmaceutical industry. Finally a methodology to determine the optimal sample size required to validate analytical methods is also given using the proposed capability metric. [less ▲]

The performance of several Internet applications often relies on the measurability of path similarity between different participants. In particular, the performance of content distribution networks mainly relies on the awareness of content sources topology information. It is commonly admitted nowadays that, in order to ensure either path redundancy or efficient content replication, topological similarities between sources is evaluated by exchanging raw traceroute data, and by a hop by hop comparison of the IP topology observed from the sources to the several hundred or thousands of destinations. In this paper, based on real data we collected, we advocate that path similarity comparisons between different Internet entities can be much simplified using lossy coding techniques, such as Bloom filters, to exchange compressed topology information. The technique we introduce to evaluate path similarity enforces both scalability and data confidentiality while maintaining a high level of accuracy. In addition, we demonstrate that our technique is scalable as it requires a small amount of active probing and is not targets dependent. [less ▲]

Echocardiography has become a routine non invasive cardiac diagnostic tool in most species. Accurate measurement of cardiac dimensions requires reference values, which are poorly documented in goats. The aim of this study was to test the inter-day repeatability and to establish the reference values of two-dimensional (2D-) and time-motion (M-) mode echocardiographic variables in healthy adult Saanen goats. Six goats were investigated three times by the same observer at one day interval using a standardized 2D- and M-mode echocardiographic protocol. The intra-observer inter-day repeatability was tested using analysis of variance, calculation of the coefficient of variation and confidence intervals. A single echocardiographic examination was performed in 6 other goats, and values obtained in the 12 goats were used to establish the 2D- and M-mode echocardiographic reference values in healthy adult female Saanen goats. Statistical analysis revealed a good inter-day repeatability of the echocardiographic cardiac measurements. Echocardiographic reference values obtained in healthy adult Saanen goats seemed slightly higher than those reported in healthy Swedish domestic goats and were similar to those reported in healthy adult sheep.
