Regulation of mRNA translation elongation impacts nascent protein synthesis and integrity and plays a critical role in disease establishment. Here, we investigate features linking regulation of codon-dependent translation elongation to protein expression and homeostasis. Using knockdown models of enzymes that catalyze the mcm(5)s(2) wobble uridine tRNA modification (U(34)-enzymes), we show that gene codon content is necessary but not sufficient to predict protein fate. While translation defects upon perturbation of U(34)-enzymes are strictly dependent on codon content, the consequences on protein output are determined by other features. Specific hydrophilic motifs cause protein aggregation and degradation upon codon-dependent translation elongation defects. Accordingly, the combination of codon content and the presence of hydrophilic motifs define the proteome whose maintenance relies on U(34)-tRNA modification. Together, these results uncover the mechanism linking wobble tRNA modification to mRNA translation and aggregation to maintain proteome homeostasis. [less ▲]

Laser-induced breakdown spectroscopy was used to analyze unburned carbon in fly ash. The samples were gotten using fast ashing method and excited by pulse laser and plasma was formed at atmospheric pressure. The emitted signal which contains the information of unburned carbon in fly ash was detected. The calibration curve constructed base on six samples of one kind of coal ash, and two samples of other two kinds of coal ash were analyzed. The experimental results show that calibration curve constructed with the data from same kind of coal ash can be used to analyze the different kinds of fly ash using proper data processing method. The relative standard deviation of repetition of measured values are less than 8.08%, the relative errors are less than 2.27%. It is indicated that laser-induced breakdown spectroscopy is suitable for rapidly detect unburned carbon in fly ash with receptible applicability. [less ▲]

The feasibility of rapid quantitative analysis of coal by Laser—Induced Breakdown Spectroscopy technique was studied.The calibration curve method for LIBS quantitative analysis is introduced.Five coal samples were selected as investigation subjects,the carbon plasma line of 505.2 nm was chosen as analysis spectrum line,and the carbon content of coal was analyzed quantitatively with delay of 0.8us, 1.2us, 1.6 us, 2.0 us and 2.4us. The results were compared with the results measured with Element Analysis Instrument.The error measured in1.6us delay was the minimum ,the effect of delay time on the quantitative analysis was analyzed.The results indicate that LIBS has a great potential in fast analysis of coal content. Samples were excited by pulse laser and plasma was formed at atmospheric pressure.The technique of laser induced breakdown spectroscopy(LIBS)was applied to analyzing the carbon content in the samples.The matrix element Si was selected as internal standard,calibration curves were drawn respectively using the peak intensity of carbon line and the intensity ratio of carbon line and silica line.The fitting degree of two different calibration curves,signal reproducibility and the accuracy of calibrated values were analyzed.The results of the analysis show that the intensity ratio method seems more adequate than the peak intensity method for establishing the calibration cures. A method of real-time evaluation of slagging propensities of coals based on element quantitative technique by laser-induced breakdown spectroscopy Was introduced .An experiment apparatus Was set up according to the principle of analysis technique,an d in which six typical coals samples were analyzed.Four ones were used to construct calibration curves of the analyzed elements as a reference diagram determining the relation between element concentration an d spectral intensity, afterwards,analyzed silicon element an d aluminum element in other sampleswere surveyed with the reference diagram .Ratios of silicon dioxide to aluminum trioxide were computed subsequently according to chemical calculation to evaluate the slagging propensities of coals.Comparing the evaluation with the computed result based on ash composition analyzed with conventional measure,the two match well.After all,some improvement program s to optimize the measurement mechanism were discussed.
