In Tunisia, to prevent and control pests and diseases during cultivation under greenhouses, chili pepper and tomato require the use of a wide range of pesticides potentially toxic and thus presenting a possible risk for farm operators, workers or consumers. A study has been carried out in the Sahel region of Tunisia to assess the risk for farm operators and workers exposed, by contact during harvest tasks, to possible pesticide residues remaining in tomato and chili pepper cultures, and for the Tunisian consumers (adults and children) after intake. A questionnaire was addressed to a group of 73 market gardeners to better understand the local professional practices and to determine the main route of exposure to pesticide. Twenty samples of cotton gloves (2 pairs / sample) were distributed to 20 volunteers who worn them for two consecutive half-days during the harvest of chili peppers or tomatoes before analysis of the dislodgeable pesticide residues which could be transferred from crops to hands. Using models predictive exposures values were calculated for consumers and farm workers. The highest exposure of consumers was observed for chlorpyrifos residues on tomatoes (with 82% and 312% of the Acute Reference Dose (ARfD), for adults and children respectively). The systemic exposure (SE) of farm workers was estimated for the average, the 90th percentile and the maximum concentration. At the highest observed concentrations, 15 pesticide residues (active substance and metabolites) used in pepper greenhouses, and 9 in tomato crops, exceed the Acceptable Operator Exposure Level (AOEL). Exposure appeared to be particularly critical for chlorothalonil sprayed in chili pepper greenhouses with SEMAX values 113 times higher than the AOEL (11285%). Long task duration (8 hours/day) after re-entry in greenhouse, limited access to personal protective equipment (PPE), lack of hygiene and bad habits (eating, drinking, or smoking at work) have also been observed and discussed as risk factors. This paper reports a novel approach to screening multi-class pesticides by ion mobility timeof- flight mass spectrometry detection. Nitrogen was selected as mobility gas. After optimization of the different ion mobility parameters, determination of matrix effect on the drift times was conducted using different matrix extracts. The results showed that drift time values are not influenced by the matrix and also are independent of the concentration within the working range for 100 pesticides tested, making drift time a powerful additional identification tool. Based on statistics, 2% variation criteria provides a good fit for all the pesticides targeted, and could be considered as a maximum acceptable criteria associated with the drift time parameter for identification purpose. This 2% value is in agreement with already reported criteria, for instance, for GC or LC retention time in European documents. Finally, the well-known feature of mobility to separate complex mixtures was also tested to obtain purified extracted mass spectra of pesticides in fruit extract. [less ▲]

Pesticide residue analysis requires methods that can determine hundreds of compounds at low levels in complex food matrices. This challenge has given rise to multi residue methods, the only efficient analytical approach. This type of analytical method entails a “generic” extraction followed by a soft or no purification step to avoid any analytes looses. With over a 1000 active compounds with different physical chemical properties, gas and liquid chromatography are used as complementary separative techniques. In the past decade, the determination has been performed on tandem mass analyzers, a powerful tool to overcome co-eluting compounds with excellent sensitivity. Nevertheless, these instruments can guarantee these results per acquisition cycles for more or less 150 compounds. This represents a serious limitation when the number of pesticides to be sought for monitoring and MRL enforcement is growing each year. As multiple injections from the same sample are not viable for laboratories, alternative options have to be explored. We propose the investigation of ion mobility (IM) coupled with mass spectrometry as a new approach for pesticide residue analysis in food. [less ▲]
